摘　　要：CML是一种发生在多能造血干细胞中的恶性骨髓增殖性肿瘤,虽然目前有多种酪氨酸激酶抑制剂可用于治疗CML,但是对于疾病复发的患者,可能会出现对后续TKI疗法产生耐药性或不耐受的问题。Asciminib是一种新型STAMP抑制剂,已于2021年10月29日经FDA受限制批准上市,用于治疗既往接受过至少2种TKI治疗的Ph+CML-CP患者或伴有T315I突变的Ph+CML-CP患者。本文对Asciminib的作用机制、药动学、药效学及最新临床研究进展和不良反应进行综述。CML is a malignant myeloproliferative tumor that occurs in pluripotent hematopoietic stem cells.Although a variety of Tyrosine Kinase Inhibitors(TKIs)are currently available for the treatment of CML,for patients with disease recurrence,subsequent TKIs may develop the problem of resistance or intolerance to therapy.Asciminib is a first-in-class STAMP inhibitor that has been approved by FDA with limited approval on October 29,2021 for the treatment of Ph+CML-CP patients who had been treated with two or more TKIs or Ph+CML in CP with the T315I mutation.This article reviews the mechanism of action,pharmacokinetics,pharmacodynamics,and the latest clinical research progress and adverse reactions of Asciminib.
