摘 要:目的研究灵芝破壁孢子粉对乳腺癌和口腔癌生长的抑制作用。方法采用灵芝破壁孢子粉乙醇提取物处理乳腺癌MCF-7细胞、口腔癌KB细胞和宫颈癌HeLa细胞,检测三种细胞的增殖情况。构建裸鼠乳腺癌MCF-7移植瘤和口腔癌KB移植瘤模型,评价灵芝破壁孢子粉进行灌胃治疗后的肿瘤生长抑制情况。结果灵芝破壁孢子粉乙醇提取物显著抑制MCF-7、KB和HeLa细胞的增殖,且该抑制效应呈剂量依赖性。在MCF-7移植瘤模型中,低、中、高3组灵芝破壁孢子粉给药剂量均有显著的肿瘤生长抑制作用,肿瘤抑制率分别为50.35%、65.72%和80.55%;但在KB移植瘤模型中,低剂量组没有显示显著的肿瘤生长抑制作用,而中剂量组和高剂量组表现出明显的抗肿瘤作用,3组肿瘤抑制率分别为4.03%、37.63%和40.59%。结论灵芝破壁孢子粉能够抑制MCF-7乳腺癌和KB口腔癌荷瘤裸鼠肿瘤的生长。OBJECTIVE To study the anti-tumor activities of sporoderm-broken Ganoderma lucidum spore powers in vitro and in vivo.METHODS Three kinds of tumor cell lines(including breast cancer cells MCF-7,oral cancer cells KB and cervical cancer cells Hela)were used to perform the in vitro studies,and they were treated with ethanol extract of sporoderm-broken Ganoderma lucidum spore powers.The cell proliferation and cell survival rate were detected by Cell Counting Kit-8(CCK-8)assay.To explore the inhibition effects of sporoderm-broken Ganoderma lucidum spore powers on tumor growth,the models of MCF-7 tumor-bearing mice and KB tumor-bearing mice were established and conducted by intragastric administration,followed by recording the mice′weight,volume and weight of tumor,and calculating the inhibition rate.Hematoxylin&eosin(H&E)and TUNEL staining were further performed to evaluate the in vivo anti-tumor activity.RESULTS The result of CCK-8 assay showed that the ethanol extract of sporoderm-broken Ganoderma lucidum spore powers possessed significant inhibition effects on cells growth of MCF-7,KB and HeLa,and it was dose-dependent.Obvious tumor growth inhibition was observed after intragastric administration at doses of 1,2 and 4 g·kg-1,and the inhibition rates were 50.35%,65.72%and 80.55%in tumor model of MCF-7,while the inhibition rates were 4.03%,37.63%and 40.59%in tumor model of KB.The results of pathological examination revealed that sporoderm-broken Ganoderma lucidum spore powers could induce tumor cell apoptosis.CONCLUSION This study suggests that sporoderm-broken Ganoderma lucidum spore powers are capable of inhibiting tumor growth on MCF-7 tumor-bearing mice and KB tumor-bearing mice.
