超声植冰协同海藻糖冷冻保存L-02肝细胞研究

摘  要:采用海藻糖代替具有生物毒性的二甲基亚砜,利用自行搭建的超声植冰装置研究了超声植冰以及不同浓度海藻糖对L-02肝细胞冷冻保存的影响。结果表明,添加0.3 mol/L海藻糖结合超声植冰的细胞冻存组的肝细胞存活率较高((87.6±2.0)%),与传统植冰组((84.3±1.0)%)无显著性差异,与非植冰组((76.9±3.1)%)存在显著性差异。Effects of ultrasonic ice seeding and different concentrations of trehalose on the cryopreservation of L-02 hepatocytes were studied by replacing bio toxic dimethyl sulfoxide with trehalose in a self-built ultrasonic ice seeding device. The results show that the survival rate of hepatocytes(87.6±2.0)% is higher when cryopreserving L-02 hepatocytes with 0.3 mol/L trehalose by ultrasonic ice seeding, which is not significantly different from the traditional ice seeding group(84.3±1.0)%, and a significantly different from the non-ice seeding group(76.9±3.1)%.
