摘　　要：目的研究藻蓝蛋白(PC)对子痫前期(PE)大鼠肝肾功能及细胞凋亡的影响。方法将50只成功受孕的Sprague-Dawley(SD)大鼠随机分为以下5组:对照组、模型组、PC低剂量组、PC中剂量组、PC高剂量组,每组10只。在孕第14天时,通过尾静脉注射1 ml脂多糖(LPS)(1μg/kg)诱导PE大鼠模型。从孕第14天到孕第20天,PC低、中、高剂量组大鼠分别灌胃50,100,200 mg/(kg·d)PC 1 ml。对照组和模型组灌胃1 ml的生理盐水。孕第21天时,检测各组大鼠收缩压(SBP)、24 h尿蛋白、谷丙转氨酶(ALT)、谷草转氨酶(AST)、血清肌酐(Scr)和血尿素氮(BUN)水平,对大鼠肝脏和肾脏进行苏木精伊红(HE)染色,采用免疫组化染色检测肝脏和肾脏中Caspase-3的表达。应用Western blot检测肝脏和肾脏中Bax、Bcl-2和cleaved Caspase-3的表达。根据试剂盒说明检测大鼠血清超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)和丙二醛(MDA)含量。采用ELISA法检测血清干扰素-γ(IFN-γ)、白细胞介素(IL)-4、IL-17和IL-10水平。结果与对照组相比,模型组SBP和24 h尿蛋白水平升高(P<0.05),其肝脏和肾脏出现损伤,肝脏和肾脏的Caspase-3染色的平均光密度(MOD)值升高(P<0.05),Bax和cleaved Caspase-3蛋白表达水平升高(P<0.05),Bcl-2降低(P<0.05)。与模型组相比,PC低、中、高剂量组SBP和24 h尿蛋白水平均降低(P<0.05),其肝脏和肾脏形态改善,肝脏和肾脏的Caspase-3 MOD值降低(P<0.05),Bax和cleaved Caspase-3蛋白表达水平降低(P<0.05),Bcl-2升高(P<0.05),且上述指标均呈PC剂量依赖性变化(P<0.05)。与对照组相比,模型组ALT、AST、Scr和BUN水平升高(P<0.05)。与模型组相比,PC低剂量组、PC中剂量组和PC高剂量组ALT、AST、Scr和BUN水平降低(P<0.05),且上述指标均呈PC剂量依赖性变化。与对照组相比,模型组血清SOD和GSH-Px水平降低,MDA升高(P<0.05)。与模型组相比,PC低剂量组、PC中剂量组和PC高剂量组血清SOD和GSH-Px水平升高,Objective To investigate the effect of phycocyanin(PC)on liver and kidney function and apoptosis in preeclampsia(PE)rats.Methods Fifty successfully conceived Sprague-Dawley(SD)rats were randomly divided into the following five groups:control group,model group,low-dose PC group,medium-dose PC group,high-dose PC group,with 10 in each group.On the 14th day of pregnancy,the PE rat model was induced by tail vein injection of 1 ml lipopolysaccharide(LPS,1μg/kg).From the 14th day to the 20th day of pregnancy,the rats in low-dose group,medium-dose group and high-dose group were given 1 ml of PC at 50,100,and 200 mg/(kg·d),respectively.Rats in control group and PE group were gavaged with 1 ml of normal saline.On the 21st day of pregnancy,systolic blood pressure(SBP),24 h urine protein,alanine aminotransferase(ALT),aspartate aminotransferase(AST),serum creatinine(Scr)and blood urea nitrogen(BUN)of rats in each group were detected.Hematoxylin eosin(HE)staining was performed on the liver and kidney of rats.The expression of Caspase-3 in liver and kidney was detected by immunohistochemical staining and the mean optical density(MOD)of Caspase-3 staining was calculated.The expression of Bax,Bcl-2 and cleaved Caspase-3 in liver and kidney was detected by Western blot.Serum superoxide dismutase(SOD),glutathione peroxidase(GSH-Px)and malondialdehyde(MDA)contents of rats were detected according to the kit instructions.Serum levels of interferon-γ(IFN-γ),interleukin(IL)-4,IL-17 and IL-10 were detected by ELISA.Results Compared with control group,SBP and 24 h urinary protein levels were increased in model group(P<0.05),liver and kidney damage were observed,Caspase-3 MOD values in liver and kidney were increased(P<0.05),Bax and cleaved Caspase-3 protein expression levels were increased(P<0.05),and Bcl-2 was decreased(P<0.05).Compared with model group,SBP and 24 h urinary protein levels were decreased in low-dose,medium-dose and high-dose groups(P<0.05),liver and kidney morphology was improved,Caspase-3 MOD values in liver and kidn
