摘 要:目的对不同产地的瓜子金药材进行质量评价。方法利用高效液相色谱-蒸发激光散射检测法(HPLC-ELSD)建立瓜子金皂苷己含量测定方法,并测定13批不同产地瓜子金药材中瓜子金皂苷己的含量,各产地样品分析采用ZORBAX Eclipse Plus C_(18)色谱柱(4.6 mm×250 mm,5μm);流动相:水-乙腈,梯度洗脱,流速:1 min/ml,柱温:30℃,进样量:10μl,漂移管温度70℃,喷雾器温度40℃,气体流速:1.64 L/min。参照2020年版《中国药典》方法对不同产地瓜子金药材进行薄层色谱鉴别。结果不同产地瓜子金药材中瓜子金皂苷含量排名:广西贵港(0.60%)>贵州毕节(0.39%)>安徽阜阳(0.37%)>贵州安顺(0.29%)>贵州兴义(0.23%)>云南双江(0.20%)>湖北随州(0.11%),其余产地未检出瓜子金皂苷己。不同产地13批瓜子金药材薄层色谱鉴别结果与高效液相色谱结果具有一致性。结论本研究建立的方法准确、科学、稳定,适用于瓜子金皂苷己的含量测定;不同产地13批瓜子金药材中仅广西贵港药材达到药典要求。Objective To evaluate the quality of Polygalae Japonicae Herba from different producing areas.Methods A method for the determination of Polygalasaponin F was established by HPLC-ELSD,and the content of Polygalasaponin F was determined in 13 batches of Polygalae Japonicae Herba from different producing areas.The samples from different producing areas were analyzed on ZORBAX Eclipse Plus C_(18)chromatographic column(4.6 mm×250 mm,5μm),with acetonitrile-water as mobile phase for gradient elution at the volume flow of 1 min/ml.The column temperature was 30℃,the injection volume was 20μl,the drift tube temperature was 70℃,the atomizing gas temperature was 40℃,and the flow rate of N_(2)in ELSD was set at 1.64 L/min.TLC identification of Polygalae Japonicae Herba from different producing areas was carried out according to the method of the Chinese Pharmacopoeia 2020 edition.Results The content of Polygalasaponin F was decreased in turn in different producing areas:Guigang in Guangxi(0.60%),Bijie in Guizhou(0.37%),Fuyang in Anhui(0.35%),Anshun in Guizhou(0.30%),Xingyi in Guizhou(0.26%),Shuangjiang in Yunnan(0.24%),Suizhou in Hubei(0.21%),and Polygalasaponin F was not detected in the other origins.The results of TLC identification and HPLC chromatography of 13 batches of Polygalae Japonicae Herba from different producing areas were consistent.Conclusion The established method is accurate,scientific and stable,which is suitable for determining Polygalasaponin F in Polygalae Japonicae Herba.Among the 13 batches of Polygalae Japonicae Herba from different producing areas,only those from Guigang in Guangxi meet the requirements of the Chinese Pharmacopoeia.
