摘  要:目的研究不同配伍比例黄连苦参对心律失常模型大鼠的治疗作用及对其心肌组织音猬因子(Shh)/胶质瘤相关癌基因同源物1(Gli1)信号通路的影响。方法120只大鼠随机分为12组,空白组、模型组、黄连:苦参3:1组、黄连:苦参2:1组、黄连:苦参3:2组、黄连:苦参1:1组、黄连:苦参2:3组、黄连:苦参1:2组、黄连:苦参1:3组、黄连组、苦参组、单煎混合液组,每组10只。空白组灌胃生理盐水10 mL·kg^(-1),其余各组灌胃等量药液。每天1次,连续预防性给药21 d。末次给药后1 h构建心律失常大鼠模型。记录各组10 min内心电图改变,统计心律失常出现时间、心律失常持续时间、室速出现时间、室速持续时间,10 s、5 min、10 min心率及10 min死亡率。实时荧光定量PCR法检测心肌组织Shh、Gli1 mRNA表达量。结果与模型组比较,不同配伍比例黄连、苦参药对水煎液组心律失常出现时间、室速出现时间延长(P<0.05),心律失常持续时间、室速持续时间缩短(P<0.05),10 s、5 min、10 min心率均增加(P<0.05)。与模型组比较,不同配伍比例黄连、苦参药对水煎液组均可降低大鼠10 min内死亡率。与模型组比较,不同配伍比例黄连、苦参药对水煎液组心肌组织Shh、Gli1 mRNA表达水平均升高(P<0.05)。以黄连:苦参2:1时效果最佳。结论黄连与苦参配伍比例为2:1时对心律失常大鼠的保护作用最强,可能通过激活Shh/Gli1信号通路发挥作用。Objective To study the therapeutic effect of golden thread and lightyellow sophora root in different proportions on the arrhythmia model rats and its effects on the Shh/Gli1 signal pathway in myocardial tissues.Methods One hundred and twenty rats were randomly divided into 12 groups,including a blank group,a model group,a coptis:sophora 3:1 group,a coptis:sophora 2:1 group,a coptis:sophora 3:2 group,a coptis:sophora 1:1 group,a coptis:sophora 2:3 group,a coptis:sophora 1:2 group,a coptis:sophora 1:3 group,a coptis group,a sophora group,and a single decoction mixture group,with 10 rats in each group.The blank group was gavaged with 10 mL·kg-1of normal saline,while the other groups were gavaged with the same amount of corresponding liquid medicines.The prophylactic administration was performed once a day for 21 days.An arrhythmia rat model was established 1 h after the last administration.The ECG changes within 10 min were recorded in each group,and the occurrence time of arrhythmia,the duration of arrhythmia,the occurrence time of tachycardia,the duration of tachycardia,the heart rate of 10 s,5 min and 10 min,and the mortality rate of 10 min were analyzed.The mRNA expression levels of Shh and Gli1 in myocardium were detected by real-time fluorescence quantitative PCR.Results Compared with the model group,the occurrence time of arrhythmia and the occurrence time of ventricular tachycardia in all the medicine groups with different compatibility ratios of coptis and sophora were prolonged(P<0.05),the duration of arrhythmia and the duration of ventricular tachycardia were shortened(P<0.05),and the heart rates were increased at 10 s,5 min and 10 min(P<0.05).Compared with the model group,coptidis and sophora of different compatibility ratios could reduce the mortality of rats within 10 min in all the medicine groups.Compared with the model group,the mRNA expression levels of Shh and Gli1 in myocardium of all the medicine groups with different compatibility ratios of coptidis and sophora were increased(P<0.05).Coptis:so
