摘  要:目的:探究不同灌注方法及灌注量对大鼠脑组织结构及神经元观察的影响并进行评价。方法:将48只wistar大鼠随机分成3组,经戊巴比妥钠麻醉后,分别采用输液器、注射器,剪右心耳或剪肝的方法进行灌注操作,比较不同灌注方法下脑组织大体形态、灌注时间及用液量的差异,并采用HE染色及免疫荧光技术对大鼠脑海马组织及神经元形态进行观察,客观化评价灌注方法选择对形态学呈现结果的影响。结果:采用输液器灌注与注射器灌注法在用时及灌注液用量方面有显著差别(P<0.01),两种方法采取右心耳引流时对大鼠脑海马组织和神经元的呈现效果基本一致,均优于剪肝引流法。结论:灌注方法的选择对大鼠脑组织及神经元固定的效果有影响,实验时长及灌注液用量对实验结果影响不显著。Objective:To explore and evaluate the effects of different perfusion methods and doses on the observation of brain structure and neurons in rats.Methods:48 Wistar rats were randomly divided into 3 groups.After sodium pentobarbital anesthesia,infusion sets,syringes,and cut right atrial appendage or liver were used for perfusion respectively.The differences of brain tissue morphology,perfusion time and liquid dosage in different perfusion directions were compared,HE staining and immunofluorescence technique were used to observe the morphology of rat hippocampal tissue and neurons,and objectively evaluate the effects of perfusion method on the morphological presentation results.Results:There were significant differences in the time and amount of perfusion fluid between infusion set perfusion and syringe perfusion(P<0.01).The two methods have basically the same effect on the hippocampus and neurons of the rat brain when the right atrial appendage is used for drainage,and both are better than the liver-cutting drainage method.Conclusion:The choice of perfusion method has an impact on the effect of rat brain tissue and neuron fixation.The length of the experiment and the amount of perfusion fluid have no significant effect on the experimental results.
