摘  要:目的 γ-氨基丁酸(γ-aminobutyric acid,GABA)是中枢神经系统重要的抑制性递质,通过观察单侧耳蜗毁损后1、2、4周GABAAα1受体亚单位在大鼠耳蜗核的表达变化,为单侧聋听觉中枢重组的分子机制提供参考。方法 新生SD(sprague-dawley)大鼠饲养至出生后12天(P12),随机选择每笼一半数量的大鼠进行单侧耳蜗毁损手术,对照组行伪手术,术后1、2、4周取实验组及对照组大鼠双侧耳蜗核,切片后采用免疫组化方法观察耳蜗核GABAAα1受体亚单位的表达变化。结果实验组术侧及健侧耳蜗核中GABAAα1受体亚单位的表达在造模后2周升高,4周降低;实验组术后2周双侧GABAAα1受体亚单位表达有显著差异(P<0.01)。实验组术后1、4周双侧耳蜗核GABAAα1表达无明显差异(P>0.05);对照组双侧耳蜗核GABAAα1表达在造模后2周升高,4周后下降,对照组大鼠术后1、2、4周双侧耳蜗核GABAAα1表达无明显差异(P>0.05);对照组术侧与实验组术侧相比术后2周有显著差异(P<0.05),术后1、4周有显著差异(P<0.01)。结论 单侧聋大鼠双侧耳蜗核GABAAα1受体的不对称改变可能为初级听觉中枢重组所必需。Objective γ-Aminobutyric acid(GABA) is an important inhibitory neurotransmitter of the central nervous system. In this study, we observed the expression of GABAAα1 subunits in rat cochlear nucleus 1, 2 and 4 weeks after unilateral cochlear lesion. Methods Newborn sprague-dawley(SD) rats were fed until 12 days after birth(P12). Half of the rats in each cage were randomly selected for unilateral cochlear destruction surgery. The control group was given sham surgery. One week, two weeks and four weeks after surgery, the bilateral cochlear nuclei of the experimental group and the control group were taken. After sectioning, the expression changes of GABAA α1 subunits in the cochlear nucleus was observed by immunohistochemistry. Results In the experimental group, the expression of GABAAα1 receptor subunits in the cochlear nucleus of the operative side and the healthy side increased 2 weeks after the model establishment, and decreased 4 weeks after the model establishment;There was a significant difference in the expression of GABAA α1 receptor subunits on both sides of the experimental group 2 weeks after operation(P<0.01).There was no significant difference in the expression of GABAA α1 in bilateral cochlear nuclei 1 week and 4 weeks after operation in the experimental group(P>0.05);In the control group, the expression of GABAAα1 in bilateral cochlear nuclei increased 2 weeks after modeling and decreased 4 weeks after modeling. In the normal control group, there was no significant difference in the expression of GABA in bilateral cochlear nuclei 1, 2 and 4 weeks after operation(P>0.05);The difference between the operation side of the control group and the operation side of the experimental group was statistically significant 2 weeks after operation(P<0.05),and the difference was statistically significant 1 week and 4 weeks after operation(P<0.01). Conclusion This experiment preliminarily suggests that the asymmetric changes of GABA receptors in bilateral cochlear nuclei of single side deafness rats,which may be
