摘  要:为了探究海南红树林/海草床沉积物中产蛋白酶细菌的多样性及产胞外酶的特性,以海南文昌红树林-海草床复合生态系统表层沉积物中的可培养产蛋白酶细菌为研究对象,通过16S rRNA测序和蛋白酶抑制剂实验进行探究.结果表明,在10个采样点共分离出111株产蛋白酶细菌,分别隶属于厚壁菌门(Firmicutes)和变形菌门(Proteobacteria),共9属,其中弧菌属菌株占总数的75.7%,为优势属.选择61株产蛋白酶活性较高的菌株,通过抑制剂实验结果显示,蛋白酶类型主要有金属蛋白酶和丝氨酸蛋白酶,占比分别为91.8%和86.9%.本研究初步探明了文昌红树林/海草床沉积物中产蛋白酶细菌的丰度及蛋白酶种类的多样性,初步探索了养殖尾水的排放对微生物群落结构的影响,这为后续开展应用性的研究奠定了基础.In the report,in order to study the diversity and extracellular enzyme activity of protease producing bacteria in the sediments of mangroves/seagrass fields in Hainan,the protease producing bacteria in the sediments of mangrove-seagrass ecosystem in Wenchang,Hainan Province in September were analyzed by 16S sequencing and protease inhibitor experiment.The results indicated that a total of 111 protease-producing bacteria are isolated from 10 sampling sites,which belong to Firmicutes and Proteobacteria.Among all nine species,Vibrio species account for 75.7%of the total.The inhibitor experiments were performed to analyze 61 strains with high protease-producing activity.The results showed that the main types of proteases are metalloprotease and serine protease,which accounted for 91.8%and 86.9%,respectively.Our findings clarified the abundance of protease-producing bacteria and the diversity of protease types in the sediments of Wenchang Mangrove-Seagrass Field,which laid an important foundation for subsequent application research.
