摘 要:建立了以有机相(甲醇)-水相(0.5%甲酸水)为流动相同时测定枣果中cAMP和cGMP含量的HPLC方法,该方法的分析条件为:Spuirsil C_(18)型色谱柱(250×4.6 mm,i.d.),流动相5%甲醇∶0.5%甲酸水=5∶95(V/V),流速0.7 mL/min,柱温30℃,检测波长256 nm。该方法具有流动相配制简单、分析时间短、色谱峰分离度好、准确度高等特点。通过10个红枣品种的分析,发现同一品种的cAMP含量高于cGMP,不同品种的cAMP和cGMP含量差异显著(p<0.05),其中骏优、骏枣和灰枣的cAMP和cGMP含量较高,这3个品种均为干制加工品种,说明cAMP和cGMP可作为新疆干制红枣品质评价的重要指标。In this paper,a HPLC method for simultaneous determination of cAMP and cGMP in jujube fruits was established by using organic phase(methanol)and aqueous phase(0.5%formic acid water)as mobile phase.Its analytical conditions were as follow:the chromatographic column was Spuirsil C_(18)(250×4.6 mm,I.D.),the mobile phase was 5%methanol∶0.5%formic acid water=5∶95(V/V),flow rate was 0.7 m L/min,the column temperature was at 30℃,and the detection wavelength was 256 nm.This method had the advantages of simple preparation of mobile phase,short analysis time,good peak separation and high accuracy.The contents of cAMP and cGMP were significantly different among different varieties(p<0.05).Junyou,Junzao and Huizao had higher contents of cAMP and cGMP,which were varieties for dried processing.These results indicated that cAMP and cGMP could be used as important indices for quality evaluation of dried jujube in Xinjiang.
