葡糖基芦丁复配物对皮肤光损伤的修护功效研究

摘  要:皮肤过度暴露于紫外线会增加ROS产生,导致红斑、色素沉着、皱纹等问题,因此,及时修复紫外线诱导的皮肤光损伤非常必要。本研究基于紫外线对皮肤的损伤机制,评估了含葡糖基芦丁、密蒙花提取物和Symrelief 100的组合物(RBS组合物)对暴露于紫外辐射下的不同皮肤细胞的作用。角质细胞实验发现,RBS组合物能够提升紫外线照射后角质细胞的活力(P<0.01),降低ROS含量(P<0.01),提升屏障相关蛋白FLG和LOR基因表达水平(P<0.01),抑制炎症因子PGE2表达(P<0.01);成纤维细胞实验发现,RBS组合物能够抑制MMP-1(P<0.05),提升I型胶原蛋白(Col I)生成(P<0.01);黑色素细胞实验发现,RBS组合物显著降低酪氨酸酶活力(P<0.01),抑制黑色素生成(P<0.05)。本研究表明,RBS组合物能够修复紫外线导致的皮肤损伤,具有改善晒伤、晒黑、晒老问题的潜力。Excessive exposure of skin to ultraviolet rays increases the production of ROS,leading to erythema,edema,pigmentation,wrinkles,sagging etc.Therefore,repairing skin photodamage induced by ultraviolet rays is necessary.In this study,the efficacy of the compositions containing glucosyl rutin,buddleja officinalis extract and Symrelief 100(RBS Combination)on different skin cells exposed to UV radiation was evaluated.Keratinocyte assay showed that RBS Combination significantly improved the viability of keratinocytes after UV irradiation,reduced the content of ROS,increased the gene expression levels of FLG and LOR,as well as inhibited the expression of proinflammatory cytokine PGE2(P<0.01).Fibroblast experiments showed that RBS Combination significantly inhibited MMP-1 synthesis(P<0.05)and promoted collagen I production(P<0.01).Melanocyte study displayed that RBS Combination significantly inhibited tyrosinase activity(P<0.01)and reduced melanin content(P<0.05).The results demonstrated that RBS Combination have the potential to repair UV-induced skin damage and ameliorate sunburn,pigmentation,and aging problems.
