摘 要:本研究拟针对浒苔(Ulva prolifera)暴发过程中微生物群落结构的变化规律进行探索,研究结果将对了解浒苔暴发期间的生物地球化学循环具有重要意义。本研究以海洋弧菌为研究对象,通过原位取样、传统培养以及高通量测序等分子生物学手段,探究青岛近岸浒苔暴发过程中弧菌丰度和群落组成的演替规律。研究结果表明,弧菌丰度随着浒苔暴发出现显著性增加,且主要发生在浒苔暴发末期。浒苔暴发过程中营养物质吸收与释放,特别是浒苔释放的溶解有机物(DOM),使弧菌群落结构出现明显演替,优势类群由浒苔暴发前的灿烂弧菌(Vibrio splendidus)转变为消退后的坎贝氏弧菌(Vibrio campbellii)。另外,温度和SiO32-是影响弧菌丰度和群落组成的重要环境因子。The outbreak of Ulva prolifera in Qingdao offshore has a serious impact on the marine ecological environment. The change law of microbial community structure during Ulva prolifera outbreak is not clear. This study will be of great significance tounderstand the biogeochemical cycle during Ulva prolifera outbreak. This study uses Vibrio as the experimental object, and through molecular biology techniques such as in situ sampling, traditional cultivable methods, and high-throughput sequencing, explores the abundance changes and succession rules of Vibrio community during the outbreak of Ulva prolifera offshore in Qingdao. The results showed that the abundance of Vibrio increased significantly with the outbreak of Ulva prolifera, and mainly occurred in the late stage of Ulva prolifera outbreak. During the outbreak of Ulva prolifera, the absorption and release of nutrients, especially the dissolved organic matter(DOM) released by Ulva prolifera, led to an obvious succession of Vibrio community structure, and the dominant group changed from Vibrio splendidus before the outbreak of Ulva prolifera to Vibrio campbellii after regression. In addition, temperature and SiO32-are important environmental factors affecting Vibrio abundance and community composition. The outbreak of Ulva prolifera caused obvious succession of Vibrio community, and temperature and silicate were important environmental control factors.
