摘 要:目的探讨枸杞多糖(LBP)对急性CO中毒后迟发性脑病(DEACMP)大鼠海马NF-κB蛋白表达的影响。方法随机将大鼠分为空白组(注射同等比例空气)、DEACMP组(改良式腹腔注射CO法建立DEACMP大鼠模型)和LBP组(40 mg/kg,连续灌胃治疗)。采用Morris水迷宫观察各组大鼠平均潜伏期,采用ELISA方法检测各组大鼠血清中IL-1β、IL-6和TNF-α表达水平,采用免疫组化观察各组大鼠脑组织中NF-κB蛋白表达情况。结果DEACMP组与LBP组染毒第14、21、28天平均潜伏期长于空白组,而LBP组大鼠平均潜伏期短于DEACMP组,差异有统计学意义(P<0.05);LBP组大鼠血清中IL-1β、IL-6和TNF-α表达水平低于DEACMP组,差异有统计学意义(P<0.05);DEACMP组与LBP组染毒第1、7、14、21、28天NF-κB蛋白表达高于空白组,且LBP组低于DEACMP组,差异有统计学差异(P<0.05)。结论LBP对DEACMP大鼠神经损伤具有很好的保护作用,其机制可能与调控NF-κB蛋白有关。Objective To investigate the effect of Lyciumbarbarum polysaccharides(LBP)on the expression of NF-κB protein in the hippocampus of rats with delayed encephalophthy after acute carbon monoxide poisoning(DEACMP).Methods The rats were randomly divided into blank group(injected with the same proportion of air),DEACMP group(modified intraperitoneal injection of CO to establish DEACMP rat model)and LBP group(40 mg/kg,continuous gavage treatment).Morris water maze was used to observe the average latency of rats in each group.The expression levels of IL-1β,IL-6 and TNF-αin serum of rats in each group were detected by ELISA.The expression of NF-κB protein in brain tissue of rats in each group was observed by immunohistochemistry.Results The average latency time of DEACMP group and LBP group on the 14th,21st and 28th day of exposure was longer than that of the blank group,while the average latency of LBP group was shorter than that of DEACMP group,the differences were statistically significant(P<0.05).The expression levels of IL-1β,IL-6 and TNF-αin the LBP group were lower than those in the DEACMP group,and the differences were statistically significant(P<0.05).The expression of NF-κB protein in DEACMP group and LBP group was higher than that in blank group on the 1st,7th,14th,21st and 28th day of exposure,and LBP group was lower than DEACMP group,the difference was statistically significant(P<0.05).Conclusion LBP has a good protective effect on nerve damage in DEACMP rats,and its mechanism may be related to the regulation of NF-κB protein.
