摘  要:目的:研究牛耳枫提取物对无水乙醇致大鼠胃溃疡的保护作用。方法:选取20只健康雄性SD大鼠进行急性毒性实验,随机分成空白组和给药组各10只,给药组在24 h内按12.5 g/kg剂量灌胃牛耳枫提取物溶液1次,空白组给予同等剂量的生理盐水。选取健康雄性SD大鼠42只,随机分成空白组、模型组、阳性药组和牛耳枫低、中、高剂量组,每组7只;阳性药组每天灌胃兰索拉唑肠溶片溶液,牛耳枫高、中、低剂量组每天分别按4、2、1 g/kg生药剂量灌胃牛耳枫提取物溶液,空白组和模型组每天给予同等剂量的生理盐水。除空白组外,其余大鼠采用无水乙醇诱导的方法建立胃溃疡模型,观察急性毒性,并测定大鼠胃溃疡面积,计算胃溃疡抑制率,观察各组大鼠胃黏膜组织形态学,测定血清中的肿瘤坏死因子-α(TNF-α)和白介素-10(IL-10)的水平,测定胃组织中丙二醛(MDA)、总抗氧化能力(T-AOC)、超氧化物歧化酶(SOD)、还原型谷胱甘肽(GSH)、谷胱甘肽过氧化物酶(GSH-Px)和非蛋白质巯基(NPSH)的含量,初探牛耳枫对胃溃疡的保护作用机制。结果:以12.5 g/kg剂量给大鼠灌胃1次,没有产生毒性,14 d内未发现死亡,14 d后解剖大鼠,脏器没有任何改变。与模型组比较,牛耳枫组胃溃疡损伤面积明显减少;从胃黏膜形态及病理组织学观察可得,牛耳枫各给药组组胃黏膜破坏显著减轻;牛耳枫各给药组组能显著性抑制TNF-α和MDA的水平,同时促进IL-10、SOD、GSH、GSH-Px和NPSH的分泌,提升T-Aoc。结论:牛耳枫提取物有很好的保护胃溃疡作用,其作用机制可能与脂质过氧化、炎症和氧化等因子调控有关。Objective:To study the protective activity of Daphniphyllum calycinum Benth.(D.calycinum)against ethanol-induced gastric ulcer in rats.Methods:20 healthy SD male rats were chosen for acute toxic experiment,which were randomly divided into the blank group and the medication group,with 10 rats in each group;the medication group was intervened with D.calycinum extract at the dose of 12.5 g/kg within 24 h,and the blank group was given the same amount of normal saline.Another 42 healthy male SD rats were randomly divided into the blank group,the model group,the positive drug group,and the D.calycinum groups of low-dose,medium-dose and high-dose,with 7 rats in each group;the positive drug group was treated with Lansoprazole Enteric-coated Tablet,the D.calycinum groups were treated with D.calycinum extract at the doses of 1 g/kg,2 g/kg and 4 g/kg,respectively,and the blank group and the model group were given the same amount of normal saline.The ethanol-induced gastric ulcer model was established.Acute toxicity was observed in rats.The area of gastric ulcer was detected,the rate of gastric ulcer inhibition was calculated,the tissue morphology of gastric mucosa was observed,the serum levels of TNF-αand IL-10were determined,and the contents of MDA,T-AOC,SOD,GSH,GSH-Px and NPSH were measured to explore the protective mechanism of D.calycinum in treating gastric ulcer.Results:There was no toxic reaction in the rats administrated with D.calycinum extract at the dose of 12.5 g/kg,and there was no death in the rats within 14 days.There were no changes in internal organs in the rats after 14 days.Compared with those in the model group,the gastric ulcer areas were significantly reduced,and the gastric mucosa damages were significantly alleviated,and the levels of TNF-αand MDA were greatly inhibited,whereas the secretions of IL-10,T-AOC,SOD,GSH,GSH-Px and NPSH were obviously promoted in the D.calycinum groups.Conclusion:D.calycinum extract has a significant protective activity against gastric ulcer induced by ethanol,which mech
