巴氏新小绥螨章鱼胺受体基因鉴定与表达模式分析

摘  要:为明确巴氏新小绥螨(Neoseiulus barkeri)章鱼胺受体基因的分子特征,探明其药理学和生理功能,运用转录组数据和生物信息学软件分析并鉴定章鱼胺受体基因的cDNA全长,利用qRT-PCR(Quantitative real-time polymerase chain reaction)比较章鱼胺受体基因在巴氏新小绥螨不同发育阶段、不同温度及饥饿胁迫条件下的表达模式.结果表明:于转录组数据库筛选鉴定到6个章鱼胺受体基因,分别命名为NbOctα1R1,NbOctα1R2,NbOctTyrR,NbOctβ2R1,NbOctβ2R2,NbOctβ3R,其完整开放阅读框为936~1 983 bp,编码312~661个氨基酸,在进化关系上与西方静走螨(Galendromus occidentalis)最为接近.NbOctα1R1,NbOctα1R2,NbOctTyrR,NbOctβ2R1,NbOctβ3R都具备G蛋白偶联受体的典型特征.6个章鱼胺受体基因在巴氏新小绥螨不同螨态均有表达,且在幼螨和若螨中表达水平较高.在不同温度胁迫条件下,不同章鱼胺受体基因在成螨和幼螨中受低温、高温影响后,仅有NbOctβ2R1,NbOctβ2R2在高温时表达量上调,低温时表达量下调.饥饿胁迫诱导时,除NbOctβ3R表达量随饥饿时间延长而降低外,其余章鱼胺受体基因表达量在饥饿开始的不同时间段出现不同程度的增加,最终随饥饿时间进一步增加而下降.从巴氏新小绥螨转录组中鉴定到的6个章鱼胺受体基因,在不同发育阶段、不同温度和饥饿胁迫下其表达量存在差异,推测巴氏新小绥螨章鱼胺受体基因在不同发育阶段和逆境胁迫中承担着不同的生理功能.In order to clarify the molecular characteristics of octopamine receptor genes of Neoseiulus barkerandexplore its physiological and pharmacologicalfunctions,bioinformaticprograms were applied to analyze the transcriptome data of N.barkeri to identify the full-length cDNA of octopamine receptor gene.Quantitative real-time polymerase chain reaction(qRT-PCR) was used to estimate the expression pattern of the octopamine receptor genes in the different developmental stages,temperatures,and starvation stress.The results show that:Six octopamine receptor genes were identified in N.barkeri and named as NbOctα1R1,NbOctα1R2,NbOctTyrR,NbOctβ2R1,NbOctβ2R2 and NbOctβ3R.Their complete open reading frames ranged from 936 to 1 983 bp.Encoding 312 to 661 amino acids,it is the closest evolutionary relation to Galendromus occidentalis.NbOctα1R1,NbOctα1R2,NbOctTyrR,NbOctβ2R1 and NbOctβ3R all possess the typical characteristics of G protein-coupled receptors.The 6 octopamine receptor genes of N.barkeri were expressed in all development stages,and the highest expression was observed in larva and nymphs.The temperature affected the expression of the octopamine receptor gene of N.barkeri,but the pattern was not obvious.The expression of NbOctβ3R gradually decreased with the increase of starvation time.The expression of other octopamine receptor genes increased in different degrees at different periods of time during the early stage of starvation,and finally decreased with the extension of starvation time.Six octopamine receptor genes were identified in N.barkeri.The expressions of octopamine receptor genes were different under different developmental stages,temperatures,and starvation stress times.This research indicated that octopamine receptor genes might play different role in different developmental stages,which can be affected by external conditions.
