摘  要:目的探究维生素D_(3)对实验性自身免疫性甲状腺炎(EAT)大鼠氧化应激的保护作用。方法建立EAT大鼠模型,设立正常对照组、模型对照组、维生素D_(3)低剂量组[0.2μg/(kg·d)]和维生素D_(3)高剂量组[2.0μg/(kg·d)],造模完成后给予维生素D_(3)治疗5周,另设维生素D_(3)预防组[2.0μg/(kg·d)]自造模开始即给予维生素D_(3)治疗,共9周。检测各组大鼠血清甲状腺过氧化物酶抗体(TPOAb)、甲状腺球蛋白抗体(TgAb)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、丙二醛(MDA)等血清指标;采用qRT-PCR法和Western blot法测定大鼠甲状腺组织中烟酰胺腺嘌呤二核苷酸磷酸(NADPH)氧化酶亚基p22^(phox)、gp91^(phox)的mRNA和蛋白表达水平。结果与正常对照组比,模型对照组TgAb、TPOAb、MDA、p22^(phox)、gp91^(phox) mRNA和蛋白表达水平均显著升高(均P<0.05),SOD、GSH-Px活性均显著降低(均P<0.05);与模型对照组比,维生素D_(3)低剂量组TgAb、MDA、gp91^(phox) mRNA和蛋白表达水平、p22^(phox) mRNA表达水平均显著降低(均P<0.05),SOD、GSH-Px活性均显著升高(均P<0.05),维生素D_(3)高剂量组和维生素D_(3)预防组TgAb、TPOAb、MDA、p22^(phox)、gp91^(phox) mRNA和蛋白表达水平均显著降低(均P<0.05),SOD、GSH-Px活性均显著升高(均P<0.05);与维生素D_(3)低剂量组比,维生素D_(3)高剂量组和维生素D_(3)预防组TgAb、TPOAb、p22^(phox) mRNA水平均显著降低(均P<0.05),维生素D_(3)预防组SOD活性显著升高(P<0.05)。结论维生素D_(3)可降低TgAb、TPOAb抗体水平和增加抗氧化应激能力,包括下调p22^(phox)、gp91^(phox) mRNA和蛋白表达水平,提高SOD、GSH-Px活性,降低MDA水平,从而发挥对EAT大鼠的保护作用。Objective To explore the effect of vitamin D_(3) on oxidative stress in experimental autoimmune thyroiditis(EAT)rats.Methods EAT was induced in rats,and the EAT rats were treated with 0μg/(kg·d)vitamin D_(3)(model control group),0.2μg/(kg·d)vitamin D_(3)(low-dose group)or 2.0μg/(kg·d)vitamin D_(3)(high-dose group)after model was established for 5 weeks,respectively;and the non-EAT rats were used as normal control group.Rats were also treated with vitamin D_(3)[2.0μg/(kg·d)]immediately after modeling initiation for 9 weeks(prevention group).Serum thyroid peroxidase antibody(TPOAb)and thyroid globulin antibody(TgAb)were measured by ELISA,superoxide dismutase(SOD)was measured by hydroxylamine method,glutathione peroxidase(GSH-Px)was measured by colorimetry and malondialdehyde(MDA)was measured by thiobarbituric acid method.The mRNA and protein expressions of NADPH oxidase subunits p22^(phox) and gp91^(phox) were determined by qRT-PCR and Western-blot,respectively.Results Compared with normal control group,serum levels of TgAb,TPOAb,MDA and the expression of expressions of p22^(phox) and gp91^(phox) mRNA and proteins were significantly increased in model control group(all P<0.05),while activities of SOD and GSH-Px were significantly decreased(all P<0.05).Compared with model control group,serum levels of TgAb,MDA and mRNA and protein expressions of gp91^(phox) and mRNA levels of p22^(phox) were significantly decreased in vitamin D_(3) treatment and prevention groups(all P<0.05),while activities of SOD and GSH-Px were significantly increased(all P<0.05).Compared with low-dose group,serum levels of TgAb,TPOAb and mRNA level of p22^(phox) high-dose group and prevention group were significantly lower(all P<0.05),while the activity of SOD in prevention group was significantly higher(P<0.05).Conclusion Vitamin D_(3) may play a protective role in EAT rats by reducing the levels of TgAb and TPOAb and increasing the antioxidant stress ability,which is associated with the down-regulated p22^(phox) and gp91^(phox) express
