摘  要:MYB基因作为植物中最庞大的一个基因家族,在花斑及色素形成、生长发育等过程中发挥着重要作用。本研究以滇水金凤花器官为材料,获得2个MYB基因,分别命名为IuMYB114和IuMYB36,其cDNA分别为315 bp和876 bp,分别编码104个和151个氨基酸。生物信息学分析显示:二者均不具有内含子且均为亲水性不稳定蛋白,IuMYB114基因属于MYB超家族,推测IuMYB36属于新的R2R3-MYB转录因子亚组;IuMYB114和IuMYB36的氨基酸序列与其他物种的同源性均在64%和50%左右;二者均分别与各自的同源序列聚在一起,且处在两个不同分支。qRT-PCR分析发现两个基因在滇水金凤斑区和非斑区中均有表达,但在斑区表达量显著高于非斑区,IuMYB114和IuMYB36基因斑区表达量分别为非斑区的12.83倍和9.88倍,推测两个基因在滇水金凤花斑形成中发挥了重要的调控作用。本研究结果为后续探讨滇水金凤花斑形成的分子调控机理以及进行凤仙花花色改良等方面研究提供了一定的理论依据。MYB genes,the largest gene family in the plant,play important roles in the formation of floral spots and pigments,as well as the growth and development of plants.In this study,with the flower of Impatiens uliginosa as material,two MYB genes named IuMYB114 and IuMYB36 were obtained by RT-PCR method,whose full cDNA lengths were 315 bp and 876 bp encoding 104 aa and 151 aa,respectively.On the basis of bioinformatics analysis,they had no introns and belonged to hydrophical and unstable proteins.The IuMYB114 gene belonged to MYB family,and IuMYB36 was speculated to be a new R2R3-MYB transcription factor subgroup.The amino acid sequences of IuMYB114 and IuMYB36 were about 64%and 50%homology with those of other species.They were clustered with their homologous sequences and located in two different branches.Based on the qRT-PCR results,they expressed in both variegation and non-variegation regions,but their expression levels in the variegation region were 12.83 and 9.88 times higher than that in non-variegation region respectively,which suggested that the two genes played important regulatory roles in the formation of variegation.These results could provide some theoretical bases for further research on the molecular regulation mechanism of flower variegation formation and color improvement of I.uliginosa.
