摘  要:目的 基于数据挖掘和网络药理学方法分析左、右半结肠癌临床组方用药规律及作用机制,为中医防治左右半结肠癌提供指导。方法 基于广州中医药大学第一附属医院2017年1月1日至2020年6月30日住院结肠癌患者施治方药信息,使用数据挖掘关联规则及聚类分析等算法,研究中医组方规律;使用网络药理学方法研究核心关联规则药物的成分、靶点及涉及的基因本体结果与京都基因和基因组百科全书(Kyoto Encyclopedia of Genes and Genomes,KEGG)通路,阐明其作用机制。结果 共纳入383首方药信息,左半结肠癌使用典型药物为48味,右半结肠癌味57味,两者共同使用典型药物44味。左半结肠癌典型方药“半枝莲-法半夏-陈皮”抗癌关键成分主要包括:麻黄素、γ-氨基丁酸及葫芦巴碱等;作用核心靶点包括:ESR1、FASLG、KITLG等;抗癌作用机制发挥涉及的主要的生物学过程包括氧水平反应、肽链内切酶活性调节及凋亡信号通路调控,涉及的信号通路主要有MAPK信号通路、RAS信号通路及PI3K-AKT信号通路。右半结肠癌典型方药“地榆-生甘草-槐花”抗癌的关键成分主要包括:白桦脂醇、延胡索乙素及熊果酸;作用核心靶点主要包括:IFNG、MAPK3、PTGS2;抗癌作用机制发挥涉及的主要的生物学过程包括:类固醇代谢过程、防御反应的正向调控及炎症反应的正向调控;涉及的通路主要包括:IL-17信号通路、VEGF信号通路及TNF信号通路。结论 中医治疗左右半结肠癌的用药规律存在一定差异。左半结肠癌方药以清热解毒、行气活血为特点,右半结肠癌则以健脾益气,养血补血为特点。左右半结肠癌的典型方药可通过多成分-多靶点-多通路的作用特点起到防治结肠癌的作用,研究结果可为中医论治左右半结肠癌的临床实践及基础研究提供参考。Objective Based on data mining and network pharmacology methods, the rules and mechanism of the clinical prescriptions were analyzed for left and right colon cancer. The present study provided the reference for the prevention and treatments of left and right colon cancer. Methods Based on the prescriptions for the colon cancer patients who admitted in the First Affiliated Hospital of Guangzhou University of Chinese drug from January 1, 2017 to June 30, 2020, the association rules algorithm and cluster algorithm were applied to analyze the rules of Traditional Chinese Medicine(TCM) prescriptions. The network pharmacology method were applied to uncover the compounds,targets, and the involved gene ontology results and KEGG pathway of the association rules. Results A total of 383 prescriptions were included. Totally, 48 typical medicines used for left colon cancer, while 57 typical medicines were used for right colon cancer, of which 44 typical medicines were used for both left and right colon cancer, for left colon cancer, the key anti-cancer compounds of the typical prescription ’Scutellaria barbata-Pinellia ternata-Chenpi’ mainly include ephedrine, γ-aminobutyric acid and trigonelline and the hub target genes includes consist of ESR1, FASLG, and KITLG. In addition, the main biological processes involved in the anti-cancer mechanism include oxygen level response,regulation of endopeptidase activity, and regulation of apoptosis signal pathways;the signal pathways involve MAPK signal pathway, RAS signal pathway and PI3K-AKT signal pathway. For left colon cancer, the key compounds of the typical prescription ’Diyu-raw licorice-Sophora japonica’ mainly include betulin, tetrahydropalmatine and ursolic acid.The hub target genes mainly include: IFNG, MAPK3, and PTGS2. The main biological processes in the anti-cancer mechanism consist of steroid metabolic process, positive regulation of defense response and positive regulation of inflammatory response.;the pathways include IL-17 signaling pathway, VEGF signaling p
