摘  要:目的探讨原因不明反复性流产(URSA)过程中微小RNA-142-3p(miR-142-3p)与细胞毒性T淋巴细胞相关蛋白4(CTLA-4)的相互作用对细胞因子影响的可能机制。方法选取2016年9月至2017年12月中国科学院大学深圳医院(光明)收治的45例URSA患者纳入流产组,选取同期于中国科学院大学深圳医院(光明)进行检查的40例健康早孕者作为对照组进行回顾性研究。采用流式细胞仪检测两组研究对象外周血中CD4^(+)CD25^(+)Treg细胞百分比,采用酶联免疫吸附法(ELISA)检测Th1型细胞因子与Th2型细胞因子的表达水平。结果流产组研究对象CD4^(+)CD25^(+)Treg细胞百分比低于对照组(P<0.05);流产组研究对象外周血中白细胞介素-10(IL-10)和白细胞介素-4(IL-4)的表达水平低于对照组(P<0.05),而白细胞介素-2(IL-2)和γ干扰素(IFN-γ)的表达水平均高于对照组(P<0.05);流产组研究对象的IL-2/IL-10、IFN-γ/IL-4比值均高于对照组(均P<0.05);荧光素酶报告基因检测显示,miR-142-3p与CTLA-4具有较强的结合能力。结论miR-142-3p对CTLA-4的表达具有潜在的抑制作用,这可能是促使URSA的机制之一。Objective To explore the possible mechanism of the interaction between microRNA-142-3p(miR-142-3p)and cytotoxic T lymphocyte-associated protein 4(CTLA-4)on cytokines during unexplained recurrent spontaneous abortion(URSA).Methods 45 female patients with URSA who were admitted to University of Chinese Academy of Sciences Shenzhen Hospital(Guangming)from September 2016 to December 2017 were selected as the abortion group,and 40 healthy early pregnant women who were examined in this hospital at the same time were selected as the control group for a retrospective study.The ratio of CD4^(+)CD25^(+)Treg cells in peripheral blood of each group was detected by flow cytometry,and the expression levels of Th1 and Th2 cytokines were detected by enzyme-linked immunosorbent assay(ELISA).Results The percentage of CD4^(+)CD25^(+)Treg cells in the control group was higher than that in the abortion group(P<0.05).Compared with the control group,the expression levels of interleukin-10(IL-10)and interleukin-4(IL-4)in peripheral blood of the abortion group were significantly decreased,while the expression levels of interleukin-2(IL-2)and interferon-γ(IFN-γ)were significantly increased(P<0.05).Meanwhile,the ratios of IL-2/IL-10 and IFN-γ/IL-4 in the abortion group were higher than those in the control group(P<0.05).Detection of luciferase reporter gene showed that miR-142-3p had strong binding ability to CTLA-4.Conclusion mi R-142-3p has a potential inhibitory effect on the expression of CTLA-4,which may be one of the possible mechanisms to promote URSA.
