摘 要:褐兜蝽(Coridius brunneus)是一种常见农林害虫,主要为害洋丝瓜和南瓜等葫芦科植物,常与药用昆虫九香虫混合发生.通过高通量测序技术,组装、拼接获得褐兜蝽线粒体基因组全序列,对其进行分析并基于13个蛋白质编码基因(PCGs)构建系统发育树.结果表明:褐兜蝽线粒体基因组的序列全长为15 792 bp(GenBank登录号:MW899158),包含13个PCGs、22个tRNAs、2个rRNAs和1个D-loop区,核苷酸组成及基因排布序列与异翅亚目昆虫一致;线粒体全序列A+T含量为75.16%,G+C含量为24.84%;预测了22个tRNAs的二级结构,除trnV缺失DHU臂外,其余均为典型的三叶草结构;系统发育分析表明兜蝽科与荔蝽科互为姐妹群关系,且兜蝽科下5个物种的系统发育关系为[(褐兜蝽C.brunneus+九香虫C.chinensis)+(短角瓜蝽Megymenum brevicorne+细角瓜蝽M.gracilicorne)]+小皱蝽Cyclopelta parva.可见,褐兜蝽与九香虫亲缘关系最近,与传统形态分类结果一致.Coridius brunneus, a common agricultural and forestry pest often occurs with medicinal insect C.chinensis, is mainly harmful to Cucurbitaceous plants such as Sechium edule and Cucurbita moschata. The mitochondrial genome of C.brunneus was obtained by high-throughput sequencing technology, and its phylogenetic tree was constructed based on 13 protein-coding genes(PCGs). The results showed that the mitochondrial genome was 15 792 bp in length(GenBank: MW899158), including 13 PCGs, 22 tRNA genes, 2 rRNA genes and 1 D-loop region. The nucleotide composition(A+T=75.16%, G+C=24.84%) and gene arrangement were consistent with those of Heteroptera insects. The secondary structures of 22 tRNAs were predicted to share the typical cloverleaf structure except for trnV which lacked an DHU arm. Phylogenetic analysis showed that Dinidoridae and Tessaratomidae were within a sister group, and the phylogenetic relationship of the 5 species under Dinidoridae was as follows: [(C.brunneus+C.chinensis)+(Megymenum brevicorne+M.gracilicorne)]+Cyclopelta parva. In conclusion, C.brunneus and C.chinensis have the closest phylogenetic relationship, which is consistent with the traditional morphological classification result.
