摘 要:‘月月粉’月季是蔷薇科蔷薇属植物,是中国古老月季品种之一,是现代月季的重要祖先.本研究基于‘月月粉’月季叶绿体基因组图谱,对其序列特征、SSR分子标记、核苷酸多态性及其在蔷薇属中的系统发育位置进行了分析.结果表明:‘月月粉’月季叶绿体基因组序列全长156 591 bp,具有非常典型的四分体结构,共有130个基因,包含86个蛋白编码基因、37个tRNA基因和8个rRNA基因,总GC含量为37.00%;‘月月粉’月季叶绿体基因组保守度极高,与其他4个近缘种‘大花香水’月季、单瓣月季、‘粉红香水’月季、‘亮叶’月季的序列有很好的共线性关系,且蛋白编码区域的保守度和相似度最高;‘月月粉’月季叶绿体基因组共有57个简单重复序列(SSR),且绝大多数是单核苷酸重复序列;‘月月粉’月季与单瓣月季的亲缘关系密切.本研究结果为后续蔷薇属的遗传多样性、遗传结构及系统发育研究提供了科学依据.As one of the ancient Chinese varieties, Rosa chinensis ‘Old Blush’ belongs to the Rosaceae family and is an important ancestor of the modern R.chinensis. In this study, the sequence characteristics, simple sequence repeat(SSR) molecular marker, nucleotide polymorphisms and phylogenetic relationship of chloroplast genome of ‘Old Blush’ were analyzed. The results showed that the 156 591 bp chloroplast genome(GC content of 37.00%) of ‘Old Blush’ had a typical quadripartite structure and contained 130 genes, including 86 protein-coding genes, 37 tRNA genes and 8 rRNA genes. The chloroplast genomes of ‘Old Blush’ was highly conserved and demonstrated certain collinearity with other 4 closely related Rosa species(R.odorata var. gigantea, R.chinensis var. spontanea, R.odorata var. erubescens and R.lucidissima), especially the protein-coding region showed the highest similarity. A total of 57 SSR sequences were identified in the chloroplast genome of ‘Old Blush’, and most of them were single nucleotide repeats. The phylogenetic analysis showed a close genetic relationship between ‘Old Blush’ and R.chinensis var. spontanea. The results provide reference for further study on the genetic diversity, genetic structure and phylogeny of Rosa.
