摘 要:近日,在福建省南平市某鸡场发现一株新的鸡传染性法氏囊病毒(IBDV)流行株,并将其命名为IBDV-FJ02.为了明晰IBDV-FJ02株遗传变异情况和临床致病特征,进行了A节段基因组序列和特征性氨基酸位点的分析、基于A节段和B节段的新型基因型分类以及对无特定病原体(SPF)鸡致病性的研究,旨在为鸡传染性法氏囊病的防治提供依据.结果显示:IBDV-FJ02株A节段核苷酸序列与早期变异株E的同源性最高,达95.6%,与中国变异株SHG558在同一分支上;IBDV-FJ02株VP2-4-3蛋白氨基酸序列第222位氨基酸为T,第249位为K,第318位为D,参考变异株中仅GLS、E与其一致,位点N^(213)、T^(222)、K^(249)、 D^(318)、E^(323)均符合变异株的特征.根据新型基因型分类方案,显示IBDV-FJ02株属于A2dB1基因型,即IBDV新型变异株类群.致病性测定结果显示:IBDV-FJ02株感染3周龄SPF鸡并不会导致感染鸡只死亡,在感染后21 d内,主要引起鸡只法氏囊的严重萎缩和脾脏的轻微肿大.上述结果表明,IBDV-FJ02株是一种IBDV新型变异株,具有明显的IBDV变异株致病特点.Recently, a new epidemic strain of Infectious Bursal Disease Virus(IBDV) was found in a chicken farm in Nanping City, Fujian Province, and it was named IBDV-FJ02. In order to clarify its genetic variation and clinical pathogenic characteristics, the genome sequence and characteristic amino acid sites of segment A, a new genotype classification based on segment A and B, and the pathogenicity of specific pathogen free(SPF) chickens were analyzed to provide a scientific basis for the prevention and treatment of infectious bursal disease in chickens. The results showed that the nucleotide sequence of segment A in IBDV-FJ02 strain had the highest homology of 95.6% with the early variant E, which was on the same branch as the variant strain SHG558 in China. The amino acid sequence of VP2-4-3 of IBDV-FJ02 strain was T at position 222, K at position 249, and D at position 318. Only GLS and E were consistent with those in the reference variant, and sites N^(213), T^(222), K^(249), D^(318), and E^(323)were consistent with the characteristics of the variant. According to the novel genotype classification scheme of IBDV, this strain belonged to the A2dB1 genotype, which was the novel variant strain group of IBDV. Pathogenicity test showed that the infection of 3-week-old SPF chickens did not result in death, within 21 d after infection, it mainly caused severe atrophy of the bursa of fabricius and slight enlargement of the spleen. In conclusion, IBDV-FJ02 was a novel variant strain of IBDV and was featured by the obvious pathogenic characteristics of IBDV variants.
