摘 要:目的:建立替代对照品法测定茯苓中7种三萜酸类成分含量。方法:色谱柱为Thermo Acclaim 120 C_(18)(4.6 mm×250 mm, 5μm),流动相为乙腈-磷酸水,梯度洗脱,流速1 mL·min^(-1),检测波长:210 nm,柱温30℃,进样量20μL。以中药标准物质替代品熊果酸作为内参物,分别建立去氢土莫酸、土莫酸、猪苓酸C、3-表去氢土莫酸、3-O-乙酰基-16α-羟基-氢化松苓酸、去氢茯苓酸、茯苓酸的相对校正因子,计算其含量,并与外标法测定结果进行对比分析,验证替代对照品法的合理性、可行性。结果:所建立的7种三萜酸相对校正因子重复性良好,25批茯苓中测定结果的计算值与实测值无显著性差异。结论:该方法测定结果可靠,实现了只用一个对照品同步测定茯苓中多个成分的方法,可为中药多指标成分质量评价模式提供参考。OBJECTIVE To establish a method for the determination of seven triterpenoids in Poria cocos.METHODS The chromatographic column was Thermo Acclaim 120 C_(18)(4.6 mm×250 mm, 5 μm), the mobile phase was acetonitrile phosphoric acid water with gradient elution at the flow rate of 1 mL·min^(-1), the detection wavelength was 210 nm, the column temperature was set at 30 ℃, and the injection volume was 20 μL. With a substitute for the standard substance of traditional Chinese medicine ursolic acid as the internal reference substance, the relative correction factors of dehydromic acid, thymic acid, polyporus acid C, 3-epidehydromic acid, 3-O-acetyl-16α-hydroxy hydropinellic acid, dehydrogenated Poria acid and poria acid were established, and their contents were calculated and compared with the results of an external standard method to verify the rationality and feasibility of the alternative reference method.RESULTS The relative correction factors of the 7 triterpenoid acids were well reproducible. There was no significant difference between the calculated and measured values of 25 batches of Poria cocos.CONCLUSION The method is reliable, and can be used for the simultaneous determination of multiple components in Poria cocos with only one reference substance. It can provide reference for the quality evaluation model of multi index components in traditional Chinese medicine.
