摘 要:为研究凉山黑绵羊在发情前期、发情期、发情后期和发情间期BMPR-IB基因不同基因型与生殖激素之间的调控关系,利用公羊试情法和外阴部观察法判断发情状态,采集凉山黑绵羊4个时期的颈静脉血,提取DNA,利用PCR扩增、酶切技术、琼脂糖凝胶电泳检测BMPR-IB基因的基因型,通过ELISA技术检测4种主要生殖激素在不同生理时期血液中的含量。结果显示,凉山黑绵羊BMPR-IB基因共有BB型、B+型和++型3种基因型。在发情期,FSH分泌量BB基因型显著高于B+基因型和++基因型(P<0.05);在发情前期,LH分泌量BB基因型显著高于B+基因型和++基因型(P<0.05);在发情中期和发情后期,P4分泌量BB基因型大于B+基因型和++基因型,且差异显著(P<0.05);E2分泌量在4个时期BMPR-IB基因不同基因型之间差异均不显著(P>0.05)。综上,凉山黑绵羊BMPR-IB存在多胎基因,而且有3种基因型;BMPR-IB基因的突变对FSH、LH和P4的分泌有促进作用。The aim of this study was to analyze the regulatory relationship between BMPR-IB different genotypes and reproductive hormones in Liangshan Black sheep during proestrus,estrus,metestrus and interestrus.Estrus state of Liangshan Black sheep was judged by the method of the ram testing and extrinsic observation.The neck venous blood was collected at four stages,and DNA was extracted.Then the BMPR-IB different genotypes were detected by PCR amplification,enzyme digestion technology and agarose gel electrophoresis.The blood levels of four major reproductive hormones in different physiological periods were detected by ELISA.The results showed that BB genotype,B+genotype and++genotype were detected in Liangshan Black sheep in this study.During the estrus period,the amount of FSH secreted by BB genotype was higher than that of B+genotype and++genotype(P<0.05).The amount of LH secreted of BB genotype was higher than that of B+genotype and++genotype(P<0.05).The secretion of P4 in BB genotype was higher than that in B+genotype and++genotype in mid and late estrus,and the difference was significant(P<0.05).But there was no significant difference in E2 content among the different genotypes of BMPR-IB gene in the four periods(P>0.05).In conclusion,there were multiple of BMPR-IB gene in Liangshan Black sheep,and had three genotypes.The mutation of BMPR-IB gene promoted the secretion of FSH,LH and P4.
