摘 要:目的探讨枸杞多糖(Lycium barbarum polysaccharides,LBP)对压疮(pressure ulcer,PU)大鼠创面愈合及Wnt/β-catenin信号通路的影响。方法采用随机数字表法将18只SD大鼠分为3组,即空白组、模型组和LBP组。采用缺血再灌注损伤方法制备PU大鼠模型并给予LBP干预2周。观察各组大鼠治疗后第1天、3天、7天和14天创面的治愈率。创面组织取材进行HE染色病理检测。采用ELISA进行血清肿瘤坏死因子(tumor necrosis factor,TNF)-α、白细胞介素(interleukin,IL)-6、IL-1β、超氧化物歧化酶(superoxide dismutase,SOD)、丙二醛(malondialdehyde,MDA)、血管内皮生长因子(vascular endothelial growth factor,VEGF)表达水平检测。Wnt1、β-catenin和GSK-3β的mRNA和蛋白表达水平采用RT-qPCR方法和蛋白印迹方法进行检测。结果在第3天、7天和14天时,LBP组大鼠创面愈合率明显高于模型组,差异有统计学意义(P<0.05)。HE染色结果表明LBP可以减轻组织炎性浸润增加毛细血管数量。LBP治疗后与模型组比较可以有效降低血清中TNF-α、IL-6和IL-1β的表达水平,差异有统计学意义(P<0.05)。LBP治疗后与模型组比较,可以明显提高血清SOD和VEGF表达水平,降低MDA表达水平,差异有统计学意义(P<0.05)。与模型组比较,LBP治疗后PU大鼠创面组织中Wnt1和β-catenin的mRNA及蛋白表达明显升高,而GSK-3β的mRNA及蛋白表达明显降低,差异有统计学意义(P<0.05)。结论LBP在PU大鼠创面愈合过程中发挥了抗氧化和抗炎的特性,同时通过对Wnt/β-catenin信号通路的调控达到促进皮肤创面愈合的作用。Objective To investigate the effect of Lycium barbarum polysaccharides(LBP)on wound healing in rats with pressure ulcer(PU)based on Wnt/β-catenin signaling pathway.Methods Eighteen SD rats were divided into 3 groups by random number table method:blank group,model group and LBP group,6 rats in each group.The PU rat model was established by ischemia-reperfusion injury and treated with LBP for 2 weeks.The healing rates of the wounds on the 1st,3rd,7th and 14th days after treatment in each group were observed.The wound tissue was sampled for pathological examination by hematoxylin-eosin(HE)staining.The expression levels of serum tumor necrosis factor-α(TNF-α),interleukin 6(IL-6),interleukin-1β(IL-1β),superoxide dismutase(SOD),malondialdehyde(MDA)and vascular endothelial growth factor(VEGF)were detected by enzyme linked immunosorbent assay(ELISA).The mRNA and protein expression levels of Wnt1,β-catenin and GSK-3βwere detected by real-time quantitative polymerase chain reaction(RT-qPCR)and Western blotting.Results The wound healing rate of the rats in the LBP group was significantly higher than that in the model group at 3days,7days and 14days,and the difference was statistically significant(P<0.05).HE staining showed that LBP reduced inflammatory infiltration and increased the number of capillaries.Compared with the model group,the expression levels of TNF-α,IL-6 and IL-1βin serum were effectively decreased after LBP treatment,and the differences were statistically significant(P<0.05).Compared with the model group,the expression levels of serum SOD and VEGF were significantly increased,while the expression level of MDA were significantly decreased after LBP treatment,the difference were statistically significant(P<0.05).Compared with the model group,the mRNA and protein expressions of Wnt1 andβ-catenin in the wound tissue of PU rats after LBP treatment were significantly increased,while the mRNA and protein expressions of GSK-3βwere significantly decreased,and the difference were statistically significant(P
