摘 要:目的探讨PYCR1在肝癌中的表达特点及其临床价值。方法运用生物信息学数据库分析PYCR1在肝癌组织中的表达及其与患者预后的关系;分别采用免疫组化和ELISA实验检测肝癌组织及患者外周血中PYCR1表达水平;应用ROC曲线评价PYCR1对肝癌的诊断价值。结果数据库分析发现,PYCR1在肝癌组织中的表达明显高于正常肝组织,PYCR1高表达肝癌患者预后较低表达患者差。肝癌组织(25/41,61.0%)PYCR1表达明显高于正常肝组织(1/25,4.0%),且其表达水平与患者年龄、性别无关。肝癌患者血清PYCR1蛋白水平明显高于肝良性疾病患者,且其表达水平与患者年龄、性别无关。PYCR1诊断肝癌的cut-off值为102.1pg/ml,曲线下面积(AUC)为0.711,敏感度为79.3%,特异性为64.7%。结论肝癌组织及患者外周血PYCR1高表达,其升高水平对肝癌具有一定的诊断及预后价值,可以作为潜在的肝癌标志物。Objective To investigate the expression characteristics and clinical value of PYCR1 in hepatocellular carcinoma(HCC).Methods Bioinformatics database was used to analyze the expression of PYCR1 in HCC and its relationship with prognosis of patients.The expression levels of PYCR1 in tissues and peripheral blood of patients with HCC were detected by immunohistochemistry and enzyme linked immunosorbent assay(ELISA),respectively.The diagnostic value of PYCR1 was evaluated by receiver operator characteristic(ROC)curve in HCC.Results Database analysis showed that the expression of PYCR1 in HCC tissues was significantly higher than that in normal liver tissues,and the prognosis of HCC patients with high expression of PYCR1 was worse than that of patients with low expression of PYCR1.The expression of PYCR1 in HCC tissues(25/41,61.0%)was significantly higher than that in normal liver tissues(1/25,4.0%),and the expression level of PYCR1 was not related to the age and gender of patients.The level of serum PYCR1 protein in patients with HCC was significantly higher than that in patients with benign liver disease,and the expression level of serum PYCR1 protein was not related to the age and gender of patients.The cut-off value of PYCR1 in the diagnosis of HCC was 102.1pg/ml,the AUC was 0.711,the sensitivity was 79.3%,and the specificity was 64.7%.Conclusion The high expression of PYCR1 in tissues and peripheral blood of patients with HCC has certain diagnostic and prognostic value for HCC,and which can be used as a potential marker of HCC.
