摘  要:目的探讨红细胞分化调节因子1(Erdr1)对小鼠玫瑰痤疮模型中辅助性T细胞1(Th1)/Th2分化的作用。方法建立玫瑰痤疮模型,随机分为模型组、重组Erdr1(rErdr1)低、高剂量组,另设对照组。评估各组小鼠皮损红斑评分,比较血清免疫球蛋白、外周血T细胞亚群水平,比较各组皮损组织中干扰素-γ(IFN-γ)、白细胞介素-10(IL-10)、IFN-γ/IL-10水平,比较各组皮损组织中Erdr1蛋白表达。结果与模型组比较,rErdr1低剂量组、rErdr1高剂量组皮损红斑评分,血清IgM、IgA、IgG,CD4^(+)T细胞百分率,皮损组织中IFN-γ、IFN-γ/IL-10水平均降低,CD8^(+)T细胞百分率、皮损组织中IL-10水平及Erdr1蛋白相对表达量均升高(P<0.05);rErdr1高剂量组上述指标改变较低剂量组更明显(P<0.05)。结论rErdr1可有效改善玫瑰痤疮小鼠皮损红斑症状,减轻炎性反应,其可能通过抑制获得性免疫应答能力及调节Th1/Th2细胞分化平衡发挥作用。Objective In order to investigate the effects of erythroid differentiation regulator 1(Erdr1)on the differentiation of Th1/Th2 in mice models of rosacea.Methods The models of rosacea were established and randomly divided into model group,recombinant erythroid differentiation regulator 1(rErdr1)low dose and high dose group,and control group.The skin lesion erythema scores of each group of mice were evaluated,the levels of serum immunoglobulin,peripheral blood T cell subsets,and interferon-γ(IFN-γ),interleukin-10(IL-10),IFN-γ/IL-10,Erdr1 protein expression in the skin lesions were compared.Results Compared with model group,the skin lesion erythema score,serum IgM,IgA,IgG,CD4^(+)cell percentage,and IFN-γ,IFN-γ/IL-10 levels were decreased,the percentage of CD8^(+)T cells,the level of IL-10 and the relative expression of Erdr1 protein were increased in rErdr1 low dose group and rErdr1 high dose group(P<0.05).The above indicators of rErdr1 high dose group were changed more significantly than rErdr1 low dose group(P<0.05).Conclusion rErdr1 can effectively improve the skin lesions and erythema symptoms of rosacea mice and reduce inflammation.It may play a role by inhibiting the adaptive immune response and regulating the balance of Th1/Th2 cell differentiation.
