摘  要:目的建立一测多评法同时测定金叶败毒颗粒中5-咖啡酰奎宁酸、绿原酸、4-咖啡酰奎宁酸、咖啡酸、芸香苷、木犀草苷、3,4-二咖啡酰奎宁酸、3,5-二咖啡酰奎宁酸、4,5-二咖啡酰奎宁酸的含量。方法该药物70%甲醇提取液的分析采用Agilent Zorbax SB C_(18)色谱柱(250 mm×4.6 mm,5μm);流动相20 mmol KH_(2)PO_(4)(磷酸调节pH至2.5)-(乙腈-甲醇,85∶15),梯度洗脱;体积流量1.0 mL/min;柱温30℃;检测波长324 nm。以绿原酸为内标,建立其他8种成分的相对校正因子,测定其含量。结果9种成分在各自范围内线性关系良好(r≥0.9997),平均加样回收率98.01%~102.91%,RSD 0.92%~2.12%,一测多评法所得结果与外标法接近。结论该方法稳定可靠,可用于金叶败毒颗粒的质量控制。AIM To establish a quantitative analysis of multi-components by single-marker(QAMS)method for the simultaneous content determination of 5-caffeoylquinic acid,chlorogenic acid,4-dicaffeoylquinic acid,caffeic acid,rutin,luteoloside,3,4-dicaffeoylquinic acid,3,5-dicaffeoylquinic acid and 4,5-dicaffeoylquinic acid in Jinye Baidu Granules.METHODS The analysis of 70%methanol extract of this drug was performed on a 30℃ thermostatic Agilent Zorbax SB C_(18)column(250 mm×4.6 mm,5μm),with the comprising of 20 mmol KH_(2)PO_(4)(pH 2.5 adjusted with phosphoric acid)-(acetonitrile-methanol,85∶15)flowing at 1.0 mL/min in a gradient elution manner,and the detection wavelength was set at 324 nm.Chlorogenic acid was taken as an internal standard to calculate the relative correction factors of the other eight constituents,after which the content determination was made.RESULTS Nine constituents showed good linear relationships within their own ranges(r≥0.9997),whose average recoveries were 98.01%-102.91% with the RSDs of 0.92%-2.12%.The results obtained by QAMS approximated those obtained by external standard method.CONCLUSION This stable and reliable method can be used for the quality control of Jinye Baidu Granules.
