摘 要:目的观察槐角苷对萎缩性阴道炎大鼠的保护作用。方法将50只大鼠随机分为假手术组、模型组、阳性药组、赋型剂组、槐角苷组,切除卵巢进行造模,假手术组和模型组不给药,其余组连续阴道给药14 d后,观察用药前后大鼠一般体征变化(体质量、阴道pH、阴道健康状况、乳酸杆菌、内糖原),血清E2和FSH水平,阴道上皮黏膜中AQP1、AQP3蛋白和mRNA表达。结果与模型组比较,槐角苷组大鼠体质量、血清E2和FSH水平无明显变化(P>0.05),阴道健康评分、乳酸杆菌数量、上皮细胞数量、内糖原表达、AQP1、AQP3蛋白和mRNA表达升高(P<0.05),炎细胞数量、阴道pH降低(P<0.05)。结论槐角苷对萎缩性阴道炎大鼠具有保护作用,其机制与提高大鼠阴道上皮黏膜AQP1和AQP3表达有关。AIM To observe the protective effects of sophoricoside on rat models of atrophic vaginitis.METHODS Fifty rats were randomly divided into the sham operation group,the model group,the positive drug group,the excipient group and the sophoricoside group,with ovariectomy performed for establishment of rat models of atrophic vaginitis.All the groups,except for the sham operation group and the model group left with no more treatment,were given 14 days consecutive application of vaginal medicine.The rats had their changes of general signs(body weight,vaginal pH,vaginal health,levels of lactobacillus and glycogen),serum E2and FSH levels,and the protein and mRNA expressions of AQP1,AQP3 in vaginal epithelial mucosa observed anteroposterior the treatment.RESULTS Compared with the model group,the sophoricoside group displayed no significant changes in body weight,serum E2and FSH levels(P>0.05);increases in vaginal health score,counts of lactobacillus and epithelial cells,glycogen expression,and protein and mRNA expressions of AQP1 and AQP3(P<0.05),decreases in the number of inflammatory cells and vaginal pH value(P<0.05).CONCLUSION The protective effects of sophoricoside on rat models of atrophic vaginitis may be related to the increased expression of AQP1 and AQP3 in vaginal epithelial mucosa.
