摘  要:目的研究滇重楼Paris polyphylla var.yunnanensis(Franch.)Hand.-Mazz.茎叶的黄酮类化学成分,并对其体外抗血小板聚集作用进行研究。方法滇重楼60%乙醇提取物采用硅胶柱色谱、ODS柱色谱、Sephadex LH-20凝胶柱色谱以及半制备型高效液相色谱进行分离纯化,根据理化性质及波谱数据鉴定所得化合物的结构。并对其中的主要化学成分进行体外抗血小板聚集活性研究。结果从中分离得到13个化合物,分别鉴定为kaempferol 7-O-β-D-glucopyranoside(1)、7-O-α-L-rhamnopyranoside-kaempferol-3-O-β-D-glucopyranoside(2)、isorhamnethin 3-O-glucoside(3)、quercetin 3-O-β-glucopyranoside(4)、kaempferol-3-O-β-robinobinoside(5)、kaempferol-3-O-α-L-rhamnopyranosyl-(1→6)-β-D-glucopyranoside(6)、isorhamnetin-3-O-rutinoside(7)、isorhamnetin-3-O-β-gentiobioside(8)、kaempferol(9)、isorhamnetin(10)、3-O-[α-L-arabmopyranosyl-(1→6)-β-D-galactopyranosyl]kaempferol(11)、isorhamnetin-3-O-β-D-galactopyranosyl-(1→6)-β-D-glucopyranoside(12)、kaempferol 3-O-β-D-glucopyranosyl-(1→3)-β-D-glucopyranoside 7-O-α-L-rhamnopyranoside(13)。化合物2、8能抑制ADP诱导的体外血小板聚集,化合物13对抑制ADP诱导的体外血小板聚集有一定的趋势。结论化合物1~6、9、11~13为首次从该植物中分离得到。滇重楼茎叶具有良好的抗血小板聚集的应用前景。AIM To study the chemical constituents from the stems and leaves of Paris polyphylla var.yunnanensis(Franch.)Hand.-Mazz.,and their anti-platelet aggregation effects in vitro.METHODS The 60%ethanol extract from P.polyphylla was isolated and purified by silica gel column chromatography(CC),ODS CC,Sephadex LH-20 gel CC and semi-preparative HPLC,then the structures of obtained compounds were identified by physicochemical properties and spectral data.In vitro anti-platelet aggregation activity of the main chemical components was studied.RESULTS Thirteen compounds were isolated and identified as kaempferol 7-O-β-D-glucopyranoside(1),7-O-α-L-rhamnopyranoside-kaempferol-3-O-β-D-glucopyranoside(2),isorhamnethin 3-O-glucoside(3),quercetin 3-O-β-glucopyranoside(4),kaempferol-3-O-β-robinobinoside(5),kaempferol-3-O-α-L-rhamnopyranosyl-(1→6)-β-D-glucopyranoside(6),isorhamnetin-3-O-rutinoside(7),isorhamnetin-3-O-β-gentiobioside(8),kaempferol(9),isorhamnetin(10),3-O-[α-L-arabmopyranosyl-(1→6)-β-D-galactopyranosyl]kaempferol(11),isorhamnetin-3-O-β-D-galactopyranosyl-(1→6)-β-D-glucopyranoside(12),kaempferol 3-O-β-D-glucopyranosyl-(1→3)-β-D-glucopyranoside 7-O-α-L-rhamnopyranoside(13).Compounds 2 and 8 can significantly inhibit ADP-induced platelet aggregation,compound 13 showed a certain tendency to inhibit ADP-induced platelet aggregation in vitro.CONCLUSION Compounds 1-6,9 and 11-13 are first isolated from this plant.the stems and leaves of P.polyphylla have good application prospects for anti-platelet aggregation.
