外翻肠囊法研究异功散的肠吸收特征

摘　　要：目的:研究异功散主要成分的肠吸收及动力学特征,判断其主要吸收成分。方法:采用大鼠离体外翻肠囊模型进行肠吸收实验,计算各成分的吸收参数。结果:6种主要成分甘草苷、橙皮苷、甘草酸、芸香柚皮苷、芹糖异甘草苷、人参皂苷Re的表观渗透系数(P_(app))为1×10^(-6)~1×10^(-5) cm·s^(-1);除甘草酸和芹糖异甘草苷外其他成分的吸收速率常数(K_(a))在不同质量浓度组间差异有统计学意义(P<0.05),且均随质量浓度呈线性增加(r>0.97)。结论:异功散中6个主要成分的小肠吸收良好,且以被动转运为主,其中人参皂苷Re的吸收最好,其次为甘草苷、甘草酸、芸香柚皮苷、芹糖异甘草苷、橙皮苷。该结果可为确定异功散的药效物质基础提供参考。Objective:To study the intestinal absorption and dynamic characteristics of the main components of Yigongsan and determine the main absorbable components.Methods:The intestinal absorption experiment was carried out by using the rat model of everted intestinal sac in vitro,and the absorption parameters of each component were calculated.Results:The apparent permeability coefficient(P_(app))values of liquiritin,hesperidin,glycyrrhizic acid,narirutin,isoliquiritin apioside,and ginsenoside Re were 1×10^(-6)-1×10^(-5) cm·s^(-1).Except those of isoliquiritin apioside and glycyrrhizic acid,the absorptionrateconstant(K_(a))values of the other four compounds existed statistical significant differences among groups with different concentrations(P<0.05),and increased linearly with concentration(r>0.97).Conclusion:The six components are well absorbed mainly through passive transportation in the intestine.The intestinal absorption of ginsenoside Re is the best,followed by liquiritin,glycyrrhizic acid,narirutin,isoliquiritin apioside,and hesperidin.The results of this study can provide references for determining the effective substances of Yigongsan.
