摘 要:目的 研究桧木醇对乳腺癌MCF-7细胞增殖和凋亡的影响及其相关机制。方法 用0、5、10和20μmol/L桧木醇处理人乳腺癌MCF-7细胞,采用划痕法检测细胞的迁移、Transwell法检测细胞侵袭能力、流式细胞技术检测细胞凋亡情况、采用蛋白免疫印迹法检测TGF-β1/Foxp3/RORγt信号通路相关蛋白的表达。结果 与对照组比较,5、10和20μmol/L桧木醇组细胞细胞划痕愈合率、侵袭能力均显著降低(P<0.01或P<0.05),且随着桧木醇浓度的升高,细胞划痕愈合率和侵袭能力逐渐下降,差异有统计学意义(P<0.05);细胞凋亡率显著升高(P<0.01或P<0.05),且随着桧木醇浓度的升高,细胞凋亡率逐渐升高,差异有统计学意义(P<0.05);TGF-β1、Foxp3、RORγt蛋白显著降低(P<0.05),随着桧木醇浓度的升高,TGF-β1、Foxp3、RORγt蛋白表达水平逐渐降低,差异有统计学意义(P<0.05)。结论 桧木醇可以通过抑制乳腺癌细胞中TGF-β1/Foxp3/RORγt信号通路来调节乳腺癌细胞的侵袭、迁移和凋亡,具有成为乳腺癌免疫治疗的潜在靶点。Objective To investigate the effects of linalool on the proliferation and apoptosis of breast cancer MCF-7 cells and its related mechanisms. Methods Human breast cancer MCF-7 cells were treated with different concentrations of lindenyl(0 μmol/L,5 μmol/L,10 μmol/L,and 20 μmol/L,respectively). Cell migration was detected by scratch assay,cell invasion ability was detected by Transwell method,flow cytometry was used to detect apoptosis,and Western blotting was used to detect the expression of TGF-β1/Foxp3/RORγt signaling pathway-related proteins. Results After the cells were treated with linalool,the cells in the linalool(5 μmol/L,10 μmol/L,and 20 μmol/L)group had a significantly reduced healing rate and invasion ability compared to those in the control group(P<0.01 or P<0.05). And with the increase of linalool concentration,the scratch healing rate and invasion ability of cells gradually decreased,and the difference was statistically significant(P<0.05). Compared with the control group,the linalool(5 μmol/L,10 μmol/L,and 20 μmol/L)group cells had a significantly higher apoptotic rate(P<0.01 or P<0.05). And with the increase of linalool concentration,the apoptosis rate gradually increased,the difference was statistically significant(P<0.05). Compared with the control group,the linalool(5 μmol/L,10 μmol/L,and 20 μmol/L)groups significantly reduced the expression of TGF-β1,Foxp3,and RORγt proteins(P<0.01 or P<0.05). With the increase of linalool,the TGF-β1,Foxp3,and RORγt protein expression levels gradually decreased,and the difference was statistically significant(P<0.05). Conclusion Linalool can regulate the invasion,migration and apoptosis of breast cancer cells by inhibiting the TGF-β1/Foxp3/RORγt signaling pathway in breast cancer cells,and has potential targets for breast cancer immunotherapy.
