扶正解毒方对脓毒血症肠损伤大鼠模型Rho/Rock信号通路的影响

摘  要:目的探讨扶正解毒方减轻脓毒血症肠损伤的作用及机制。方法将60只大鼠随机分为假手术组、模型组、Rho激酶抑制剂组(Y-27632)、扶正解毒方低剂量组、扶正解毒方高剂量组、蒙脱石散组,每组各10只。药物干预4周后,收集大鼠血清及结肠组织,HE染色观察各组大鼠结肠组织形态学改变,ELISA法比较各组大鼠血清炎症因子水平、氧化应激水平,Western blottng法检测各组大鼠结肠组织Rho/Rock信号通路相关蛋白水平。结果低剂量、高剂量扶正解毒方均可以明显改善大鼠脓毒血症肠损伤,有效降低大鼠血清肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)、HMBG1水平(P<0.05),并且血清及结肠组织丙二醛(MDA)、超氧化物歧化酶(SOD)的含量也明显下降(P<0.05),扶正解毒方低剂量、高剂量组大鼠结肠组织Rho、Rock蛋白的表达显著下调(P<0.05),ZO-1、Occludin蛋白的表达显著上调(P<0.05)。结论扶正解毒方可以通过抑制Rho/Rock信号通路,减少肠道炎症和氧化应激,从而改善大鼠脓毒血症引起的肠道损伤,其作用呈一定的剂量依赖性。Objective:To investigate the effect and mechanism of Fuzheng Jiedu Decoction in alleviating septic intestinal injury.Methods:A total of 60 rats were randomly divided into the sham operation group,model group,Rho kinase inhibitor Y-27632 group,low-dose Fuzheng Jiedu Decoction group,high-dose Fuzheng Jiedu Decoction group,and Mengtuoshisan group,with 10 rats in each group.After four weeks of drug intervention,the serum and intestinal tissues of rats were collected,HE staining was used to observe the morphological changes of colon in each group,nd the levels of serum inflammatory factors and oxidative stress were compared by ELISA.The levels of Rho/Rock signaling pathway related proteins were detected by Western blotting.Results:Both low-dose and high-dose Fuzheng Jiedu Decoction could significantly improve the intestinal injury in rats with sepsis.The levels of TNF-α,IL-6 and HMBG1 in serum of Fuzheng Jiedu Decoction groups were significantly decreased(P<0.05),and the contents of MDA and SOD in serum and intestinal tissues were also significantly decreased(P<0.05).The expressions of Rho and Rock proteins in intestinal tissues of Fuzheng Jiedu Decoction groups were significantly decreased(P<0.05).The expression of ZO-1 and Occludin were significantly up-regulated(P<0.05).Conclusion:Fuzheng Jiedu Decoction can reduce intestinal inflammation and oxidative stress by inhibiting Rho/Rock signaling pathway,thus improve the intestinal injury caused by sepsis in rats,and the effect is dose-dependent.
