摘  要:目的观察717解毒合剂对蝮蛇咬伤大鼠心肌一氧化氮(NO)、丙二醛(MDA)、巯基(SH)水平的改善作用。方法8周龄SPF级SD大鼠50只,雌雄各半,随机分为正常对照组、模型组、717解毒合剂低剂量组(0.6 mL/100 g)、717解毒合剂中剂量组(1.2 mL/100 g)、717解毒合剂高剂量组(2.4 mL/100 g)。除正常对照组外,其余大鼠均予2.5μg/g腹蛇毒液于左后肢皮下注射造模,并于造模成功2 h后开始给药,连续3 d。末次给药12 h后取大鼠心脏制备10%心肌组织匀浆,离心后提取上清并检测心肌MDA水平、心肌NO水平、心肌总巯基(TSH)水平、心肌非蛋白结合巯基(NPSH)水平和心肌蛋白结合巯基(PSH)水平。结果与正常对照组相比,模型组大鼠心肌MDA水平和NO水平表现出显著升高的状态(P<0.05),心肌TSH、NPSH、PSH水平则呈现降低的状态(P<0.05);中药治疗组大鼠心肌MDA水平和NO水平显著下调(P<0.05),心肌TSH、NPSH、PSH水平明显上调(P<0.05),并呈现剂量依赖性趋势,给药剂量越高,心肌损伤的改善程度越优。结论717解毒合剂对蝮蛇毒诱导的大鼠心肌损害具有一定的改善和保护作用,可以改善心肌MDA、NO、TSH、NPSH、PSH水平,值得进一步研究。Objective:To observe the effect of 717 Antidote Mixture on the levels of NO,MDA and SH in myocardium in rats bitten by Agkistrodon.Methods:A total of 508-week-old SPF SD rats,half male and half,were randomly divided into normal control group,model control group,717 Antidote Mixture low-dose group(0.6 mL/100 g),717 Antidote Mixture medium-dose group(1.2 mL/100 g),717 Antidote Mixture high-dose group(2.4 mL/100 g).Except for the normal control group,the remaining rats were injected into the left hind limb with a dose of 2.5μg/g adder venene subcutaneously to establish the model.At 2 hours after successful model establishment,the administration was started and continued for 3 days.At 12 hours after the last administration,the rat heart was taken to prepare 10%myocardial tissue homogenate,the supernatant was extracted after centrifugation and the myocardial MDA level,myocardial TSH level,myocardial NPSH level,myocardial PSH level and myocardial NO level were measured.Results:Compared with the normal control group,the myocardial MDA level and NO level of the model control group showed a significantly increased state(P<0.05),while the myocardial TSH,NPSH and PSH levels showed a decreased state(P<0.05);the levels of myocardial MDA and NO of rats in the traditional Chinese medicine treatment group were significantly down-regulated(P<0.05),and the levels of myocardial TSH and NPSH were also significantly up-regulated(P<0.05)and showed a dose-dependent trend.The higher the concentration of administration,the degree of improvement of myocardial injury was better.Conclusion:717 Antidote Mixture has a certain amelioration and protection effect on myocardial damage induced by Agkistrodon venom.It can improve the levels of myocardial MDA,NO,NPSH and TSH,which is worthy of further study.
