五红汤对环磷酰胺所致免疫低下小鼠免疫功能的影响

摘　　要：目的:探究中医食疗五红汤对环磷酰胺引起的小鼠免疫功能降低的改善作用,为五红汤提高免疫力和缓解骨髓抑制提供实验依据。方法:32只C57BL/6小鼠随机分为4组,正常+五红汤组、正常对照组、模型组和五红汤治疗组,每组8只。正常+五红汤组和五红汤治疗组实验期间以五红汤作为日常饮水,其余两组饮用清洁水,连续给药19 d。实验结束后处死小鼠,计算免疫器官指数,检测血常规,ELISA法检测血清中IgG、IgM、IL-3的浓度,HE观察脾脏结构变化,免疫组化法观察STAT3、p-STAT3,流式细胞术检测脾细胞中NK细胞、CD4^(+)T及CD8^(+)T细胞比例。结果:与正常对照组相比,模型组小鼠脾脏结构损伤,STAT3表达无变化,其他各项检测指标均明显降低(P<0.05);与模型组相比,五红汤治疗组小鼠脾脏结构趋于正常,白细胞、血小板、血清IgG、IL-3水平升高(P<0.05),NK细胞、CD4^(+)T和CD8^(+)T细胞比例增高(P<0.05),p-STAT3表达明显增加(P<0.01)。结论:五红汤可用于缓解环磷酰胺导致的血细胞减少,增强免疫功能,可能是通过升高IL-3激活脾淋巴细胞STAT3的表达实现的。Objective:To investigate improving effect of Wuhong Decoction on immune function of mice induced by cyclophosphamide(CTX),and to provide experimental basis for Wuhong Decoction in improving immunity and relieving bone marrow suppression.Methods:32 C57BL/6 mice were randomly divided into the normal group,the normal plus Wuhong Decoction group,the model group and the Wuhong Decoction group,with 8 mice in each group.In normal plus Wuhong Decoction group and the Wuhong Decoction group,Wuhong Decoction was taken as daily drinking water;and in the other two groups,clear water was taken as daily drinking water.After 19 days of administration,the immune organ index was calculated,and the blood routine was detected.The serum concentrations of IgG,IgM and IL-3 were detected by ELISA method.Spleen structure changes were observed by HE staining.The contents of STAT3 and p-STAT3 were observed by immunohistochemistry.The percentage of NK cells,CD4^(+)T/CD8^(+)T were detected by flow cytometry.Results:Compared with those in the normal group,the spleen structure of mice was damaged,the expression of STAT3 did not change,and other indicators were significantly decreased in the model group(P<0.05).Compared with those in the model group,the spleen structure of mice tended to be normal,the levels of leukocytes,platelets,serum IgG and IL-3 increased(P<0.05),the percentage of NK cells and CD4^(+)T/CD8^(+)T increased(P<0.05),and the expression of p-STAT3 increased as well in the Wuhong Decoction group(P<0.01).Conclusion:Wuhong Decoction can improve cytopenia induced by CTX and enhance immune function.This may be achieved by elevating IL-3 to activate the expression of splenic lymphocyte of STAT3.
