摘 要:目的:通过观察二至丸活性部位对D-半乳糖衰老模型大鼠各组织中氧化应激相关指标及衰老蛋白的影响,探讨其延缓衰老的作用机制。方法:对二至丸中女贞子和墨旱莲进行分离提取,提取出总三萜、总黄酮及总多糖活性部位,并使用分光光度法进行活性部位测定。将70只雄性SD大鼠按体质量随机分为空白组、模型组、三萜组、黄酮组、多糖组、活性部位合并组及二至丸水煎液组共7组,以100 mg/(kg·d)的剂量腹腔注射D-半乳糖建立大鼠衰老模型,连续8周;造模后各组给药干预,连续6周。采用酶联免疫吸附法(ELISA)测定各组大鼠肝组织SOD、MDA含量,脑组织MAO、LPF含量及脑海马区P16、P21蛋白表达水平。结果:与模型组相比,各给药组大鼠SOD活性均显著升高(P<0.05,P<0.01),其中黄酮组差异无统计学意义(P>0.05);各给药组大鼠MDA、MAO、LPF含量均显著降低(P<0.01);各给药组大鼠P16、P21蛋白水平均明显下降(P<0.05,P<0.01)。与水煎液组相比,合并给药组MAO含量差异有统计学意义(P<0.05),三萜组与合并给药组P16蛋白表达差异有统计学意义(P<0.05)。结论:二至丸活性部位与水煎液的抗衰老效果相当,其中调节单胺氧化酶、P16蛋白方面,活性部位合并给药组的效果优于水煎液组。二至丸各活性部位均能够不同程度地延缓模型大鼠的衰老程度,其作用机制可能与调控脑海马区衰老蛋白表达,抑制机体氧化应激水平相关。Objective:To explore the action mechanism of Erzhi Pill in delaying aging by observing the effects of its active parts on the indexes related to oxidative stress and senescence related proteins in the aging rats induced by D-galactose.Methods:The active parts of total triterpenoids,total flavonoids and total polysaccharides were extracted from Ligustri Lucidi Fructus and Ecliptae Herba in Erzhi Pill,and the active parts were determined by spectrophotometry.70 male SD rats were randomly divided into 7 groups according to body weight,including the blank group,the model group,the triterpene group,the flavone group,the polysaccharide group,the active part combination group and the Erzhi Pill Decoction group.The aging model was established by intraperitoneal injection of D-galactose at the dose of 100 mg/(kg·d)for 8 weeks.After modeling,each group was given drug intervention for 6 weeks.ELISA method was used to determine the contents of SOD and MDA in liver tissues,MAO and LPF in brain tissues and the protein expressions of P16 and P21 in hippocampus.Results:Compared with those in the model group,the SOD activity was significantly increased in all medication groups except the flavone group(P<0.01),there was no significant difference in the flavone group(P>0.05);the contents of MDA,MAO and LPF were significantly decreased in all medication groups(P<0.01,P<0.05);the protein levels of P16 and P21 were significantly decreased in all medication groups(P<0.01,P<0.05).Compared with that in the decoction group,the content of MAO was significantly different in the combined administration group(P<0.05);the expression of P16 protein in the triterpenoid group was significantly different from that in the combined administration group(P<0.05).Conclusion:The anti-aging effect of the active parts of Erzhi Pill was equivalent to that of the decoction,and the effects of the active parts combined with the decoction group were better than those of the decoction group in terms of regulating monoamine oxidase and P16 protein.All active pa
