CBA法检测成人血清sCD25浓度及正常参考值范围

摘　　要：目的应用流式微球分析(CBA)法测定表观健康成年人血清中可溶性白细胞介素2受体(sCD25)浓度,初步建立应用CBA法检测sCD25的正常参考值范围。方法按照《临床实验室检验项目参考区间的制定》和《临床化学检验血液标本的收集与处理》标准,筛选血常规和生化指标在正常参考区间内的健康体检成年人(24~65岁)血清样本204例。根据年龄和性别分成两组,其中青年组(24~40岁)、中年组(41~65岁)各102例,两组男性、女性各51例。应用CBA方法检测各标本血清中sCD25水平,采用x±1.96s方法计算双侧95%的正常参考值范围。结果青年组和中年组血清中sCD25浓度比较差异无统计学意义(P>0.05),相同年龄段不同性别健康成人血清中sCD25浓度比较差异无统计学意义(P>0.05)。204例健康成人血清中sCD25浓度呈正态分布,平均值为(812.93±214.32)ng/L;双侧95%正常参考值范围393.87~1233.00 ng/L。结论本研究应用CBA方法初步建立了人血清sCD25浓度正常参考值,为sCD25指标在临床相关疾病方面的应用奠定了基础。Objective To determine the concentration of soluble interleukin-2 receptor(sCD25) in the serum of healthy adults by cytometric bead array(CBA), and to preliminarily establish the normal reference interval of sCD25. Methods Accor-ding to the standards of “Formulation of Reference Interval of Clinical Laboratory Test Items” and “Collection and Processing of Blood Samples for Clinical Chemical Test”, 204 serum samples of healthy adults(24-65 years old) whose blood routine and biochemical indices were within the normal reference interval were screened. They were divided into two groups according to age and sex, including 102 subjects in 24-40 years group(youth group) and 102 subjects in 41-65 years group(middle age group), with 51 males and 51 females in each group. The CBA method was used to measure serum sCD25 concentration, and the x±1.96s method was used to calculate the 95% normal reference interval on both sides. Results There was no significant difference in serum sCD25 concentration between the youth group and the middle age group(P>0.05) and between males and females in the same age group(P>0.05). The serum sCD25 concentrations of 204 healthy adults were normally distributed, with a mean of(812.93±214.32) ng/L, and the bilateral 95% normal reference interval was 393.87-1 233.00 ng/L. Conclusion In this study, the CBA method is used to preliminarily establish a normal reference value of human serum sCD25, which lays a foundation for the application of sCD25 index in clinical related diseases.
