摘 要:目的建立柴胡-白芍药对水煎液的质量标准。方法高效液相色谱(HPLC)法对柴胡-白芍药进行含量测定,色谱柱为Diamonsil(4.6nm×250nm,5μm),流动相为乙腈-0.1%磷酸水,检测波长为210nm,流速为1.0mL·min^(-1),进样量为10μL。采用上述色谱条件对13批样本进行指纹图谱分析,共标定了10个共有峰,经与对照品比对,指认了其中3个色谱峰,并同步进行含量测定。结果指认的3个色谱峰分别为芍药苷、芍药内酯苷、柴胡皂苷a,平均含量测定结果为1.40%、0.80%、0.1%。计算13批柴胡-白芍药对水煎液冻干物的色谱图相似度介于0.485~1.000。结论所建立方法操作简便、结果可靠,可用于柴胡-白芍药对水煎液定量分析,该研究为评价该药对水煎液的质量提供依据。OBJECTIVE To establish the quality standard of Bupleurum white peony decoction.METHODS The content of Radix Bupleuri and Radix Paeoniae Alba was determined by high performance liquid chromatography(HPLC).The chromatographic column was diamonsil(4.6nm×250nm,5μm)The mobile phase is acetonitrile-0.1%phosphoric acid water,the detection wavelength is 210nm,the flow rate is 1.0mL·min^(-1),and the injection volume is10μL。The above chromatographic conditions were used to analyze the fingerprint of 13 batches of samples.A total of 10 common peaks were calibrated.Three chromatographic peaks were identified by comparison with the reference substance,and the content was determined simultaneously.RESULTS The three identified chromatographic peaks were paeoniflorin,albiflorin and saikosaponin a,and the average contents were 1.40%,0.80%and 0.1%.The similarity of chromatograms of 13 batches of Bupleurum and white peony on freeze-dried matter of decoction was calculated,which was between 0.485~1.000.CONCLUSION The established method is simple and reliable,and can be used for the quantitative analysis of Bupleurum white peony decoction.This study provides a basis for evaluating the quality of Bupleurum white peony decoction.
