摘 要:采用荧光进行生物体内微塑料的生物毒性效应评价是较为常用的研究方法。选择适当的消解剂,既完全消解生物组织,又不影响荧光测定,对提高微塑料检测的可靠性具有重要的意义。以荧光聚苯乙烯微球为研究对象,选用4种不同的消解剂,分别为KOH(10(wt)%),H_(2)O_(2)(30(wt)%),HNO_(3)(35(wt)%),30%H_(2)O_(2)+35%HNO_(3)(1∶3体积比),通过对比不同消解剂对生物体和聚苯乙烯微球荧光强度的影响,选出适合的生物体内微塑料分析方法。实验结果表明,相较于其它3种消解剂,10%KOH(50℃,24h),可完全消解生物组织,对荧光微塑料的荧光强度影响最小。利用KOH消解方法对生物样品内微塑料的测定影响进行回收率评价,荧光微球回收率达到(95.2±0.5)%。It is a common method to evaluate the biotoxicity effect of microplastics in organisms using the fluorescence intensity.In order to improve the detection accuracy for microplastic,a optimal digestive agent is selected,which can completely dissolve biological tissue without affecting the fluorescence intensity.In this study,fluorescent polystyrene microplastics was taken as a research object and four commonly used digestive agents were studied:10%KOH,30%H_(2)O_(2),35%HNO_(3)and 30%H_(2)O_(2)+35%HNO_(3)(1∶3).The effect of different digestion methods on the fluorescence intensity of microplastics and organism was studied and the suitable method was selected.The results showed that,compared with the other three digestion methods,10%KOH(50℃,24h)could completely dissolve biological tissues,and had the least influence on the fluorescence intensity of the microplastics.The KOH digestion method was used to evaluate the determination of microplastics in biological samples.The recovery rate was 95.2%±0.5%.
