摘  要:目的通过体外实验观察扶正祛邪方对结肠癌细胞增殖、侵袭的抑制作用及机制。方法采用低、中、高浓度扶正祛邪方含药血清对结肠癌细胞SW480和LoVo进行干预,CCK-8和克隆形成检测其对结肠癌细胞增殖和克隆形成能力的影响,Transwell侵袭实验检测其对结肠癌细胞侵袭能力的影响,RT-qPCR检测其对长链非编码肺腺癌转移相关转录本1(MALAT1),缺氧诱导因子-1α(HIF-1α)和整合素αv(ITGαv)mRNA表达的影响,Western-Blot检测其对HIF-1α和ITGαv蛋白表达的影响。结果与对照组相比,低、中、高浓度组在48h和72h时450nm处吸光度A值较对照组显著降低(P<0.01);中、高浓度组结肠癌细胞的克隆形成数显著降低(P<0.01);低、中、高浓度组穿膜细胞数显著减少(<0.01)。不同浓度含药血清干预结肠癌细胞,均能下调MALAT1、HIF-1α和ITGαv在RNA水平的表达;与对照组相比差异显著(均<0.01)。Western-Blot结果显示,中、高浓度组HIF-1α和ITGαv蛋白表达显著下调(均<0.01)。结论扶正祛邪方能够抑制结肠癌细胞的增殖和侵袭能力,其机制可能与抑制MALAT1/HIF-1α/ITGαv信号轴有关。Objective To observe the inhibitory effect and mechanism of Fuzheng Quxie Prescription on colon cancer cell proliferation and invasion in vitro experiments.Methods Low,medium and high concentrations of Fuzheng Quxie Prescription-medical serum were used to intervene on colon cancer cells SW480 and LoVo.CCK-8 and colony formation were used to detect their effects on cell proliferation and colony formation.Transwell invasion assay was used to detect their effects on cell invasion ability of colon cancer cells.RT-qPCR was used to detect the mR NA expression of long non-coding RNAs MALAT1,HIF-1αand ITGαv,and the protein expression of HIF-1αand ITGαv was detected by Western Blot.Results Compared with control group,the absorbance A values at 450 nm in low,medium and high concentration groups at 48 h and 72h were significantly lower than those in control group(P<0.01);the number of colonies formed in low,medium,and high concentration groups was significantly decreased(P<0.01);different concentrations of drug-containing serum intervened in colon cancer cells could down-regulate the expressions of RNA of MALAT1,HIF-1αand ITGαv compared with control group(all<0.01).Results Western Blot showed that the expressions of HIF-1αand ITGαv proteins were significantly down-regulated in middle and high concentration groups(P<0.01).Conclusion Fuzheng Quxie Prescription can inhibit the proliferation and invasion of colon cancer cells,as well as the MALAT1/HIF-1α/ITGαv signaling axis.
