鸡鲍氏志贺菌ipaH基因的克隆与序列分析

摘  要:利用PCR技术扩增鸡鲍氏志贺菌侵袭性质粒抗原H基因(invasive plasmid,ipaH),并与GenBank数据库中不同菌株进行序列比对与同源性分析。结果显示,分别扩增出ipaH_1、ipaH_2、ipaH_3、ipaH_4、ipaH_5和ipaH_6等目的基因。核苷酸序列比对和同源性分析结果显示,鸡鲍氏志贺菌6个ipaH基因片段均与美国鲍氏志贺菌CP001063的核苷酸序列同源性最高,其次是中国鲍氏志贺菌CP000036,提示鸡鲍氏志贺菌可能起源于人鲍氏志贺菌。Based on PCR technology,chicken ipaH genes of Shigella boydii were cloned and sequenced,followed by analysis of sequence homology among the obtained six ipaH gene fragments.The results showed that ipaH_1,ipaH_2,ipaH_3,ipaH_4,ipaH_5 and ipaH_6 target genes were amplified respectively.The results of nucleotide sequence alignment and homology analysis showed that the 6 ipaH gene fragments of Shigella botelli of chicken had the highest nucleotide sequence homology with CP001063 of Shigella botelli of America,followed by CP000036 of Shigella botelli of China,suggesting that Shigella botelli of chicken might originate from Shigella botelli of human.
