搜风祛痰中药对动脉粥样硬化ApoE基因敲除小鼠IL-1β及IL-18的影响

摘　　要：目的探究搜风祛痰中药对动脉粥样硬化的治疗机制。方法将完成造模程序的ApoE基因敲除小鼠随机分为5大组别,包括模型组,中药低、中、高剂量组和西药组;空白对照组选用C57BL/6小鼠。对照组和模型组予生理盐水灌胃,中药组和西药组分别予不同剂量的搜风祛痰中药及阿托伐他汀干预。HE染色观察斑块面积,Western Blot法检测IL-1β及IL-18水平。结果模型组较对照组斑块面积增大,且IL-1β以及IL-18指标均有一定的提高(P<0.05);中药各组及西药组较模型组斑块面积缩小,且IL-1β以及IL-18指标均有一定的降低(P<0.05);中药高剂量组与西药组IL-1β及IL-18指标差异无统计学意义(P>0.05)。结论搜风祛痰中药可以实现IL-1β及IL-18的下调效果,缩小斑块面积,这或许为其治疗动脉粥样硬化的机制之一。Objective To explore the therapeutic mechanism of traditional Chinese medicine for Soufeng Qutan Chinese medicine on atherosclerosis.Methods ApoE gene knock-out mice that completed the modeling procedure were stochasticly divided into five groups,including model group,single,double and fourfold dosage groups of traditional Chinese medicine and western medicine group.C57bl/6 mice were taken as blank control group.By gavage the control group and the model group were fed physiological saline,traditional Chinese medicine and the western medicine group were treated with single,double and fourfold dosage of Soufeng Qutan Chinese medicine and atorvastatin respectively.Plaque area was observed by HE staining,the standards of IL-1βand IL-18 were tested by western blot.Results The area of abdominal aortic plaque with model group increased larger than that of the control group,IL-1βand IL-18 improved(P<0.05).The area of abdominal aortic plaque with traditional Chinese medicine group and western medicine group reduce smaller than that of model group,and the expression levels of IL-1β、IL-18 decreased(P<0.05).There is no statistical difference about IL-1βand IL-18 between fourfold dosage groups of traditional Chinese medicine group and western medicine group(P>0.05).Conclusion Soufeng Qutan Chinese medicine can reduce IL-1βAnd IL-18,and reduce the plaque area,which may be one of the mechanisms for its treatment of atherosclerosis.
