摘  要:为了查明引起海口市某进境实验动物隔离场3例恒河猴死亡的原因,本试验分别采集3只恒河猴的心脏、肝脏、脾脏、肺脏和淋巴结样本,进行细菌分离,通过细菌形态特征、显微镜镜检、生化鉴定、16S核糖体RNA(16S rRNA)序列分析对分离菌进行病原学鉴定;采用纸片扩散法(K-B法)测定分离菌的耐药表型;采用玻片凝集法测定分离菌的血清型;采用动物回归试验分析分离菌的致病性。结果显示,从3只恒河猴上各分离鉴定出1株大肠杆菌,命名为H01株(分离自脾脏)、H02株(分离自淋巴结)和H03株(分离自肝脏);3株大肠杆菌对12种常用抗菌药物产生不同程度耐药,并且都呈现多重耐药,其中对阿莫西林、庆大霉素、氯霉素、氟苯尼考和复方新诺明表现为耐药,仅对阿莫西林-克拉维酸钾、阿米卡星和多黏菌素B表现敏感;玻片凝集试验显示,H01株的血清型为O8,H02株的血清型为O3,H03株的血清型为O9;动物回归试验显示,3株分离菌都是强毒株。本试验为实验动物恒河猴源大肠杆菌病的防治提供参考依据。In order to find out the cause of death of three rhesus monkeys in an imported laboratory animal isolation farm in Haikou City,this study aseptically collected samples from the heart,liver,spleen,lung and lymph nodes of three rhesus monkeys for bacterial isolation followed by morphological characterization,microscopic examination,biochemical test,and 16S rRNA sequence analysis.The drug resistance phenotype of the isolated bacteria was assessed by K-B drug sensitive paper method,the serotype of the isolated bacteria was determined by slide agglutination method,and the pathogenicity of the isolates was analyzed by animal regression experiment.The results showed that three Escherichia coli strains were isolated and identified,named H01 strain(isolated from spleen),H02 strain(isolated from lymph node)and H03 strain(isolated from liver);these E.coli strains were resistant to 12 commonly used antibiotics in varying degrees,and all exhibited multiple drug resistance,including complete resistance to amoxicillin,gentamicin,chloramphenicol,florfenicol and cotrimoxazole,but complete sensitivity to amoxicillin-clavulanate potassium,amikacin and polymyxin B.Slide agglutination test showed that H01 was O8,H02 was O3,and H03 was O9;animal regression test showed that the three isolates were highly virulent.This study provides a reference for the treatment of experimental animals with rhesus monkey colibacillosis.
