摘 要:目的探讨穴位局部嘌呤物质在预电针心俞穴改善大鼠急性心肌缺血中的可能作用。方法选用雄性清洁级SD大鼠18只,随机分成生理盐水对照组、急性心肌缺血组、预电针心俞穴组,每组6只。急性心肌缺血组用背部皮下多点位注射盐酸异丙肾上腺素(ISO)制备急性心肌缺血大鼠模型,预电针心俞穴组在造模前电针双侧心俞穴3 d。全自动生化分析仪测定血清肌酸激酶同工酶(CK-MB)和超氧化物歧化酶(SOD)活性;酶联免疫吸附试验(ELISA)测定心俞穴穴区组织中嘌呤物质三磷酸腺苷(ATP)、二磷酸腺苷(ADP)和一磷酸腺苷(AMP)浓度。结果与生理盐水对照组比较,急性心肌缺血组血清SOD显著下降(P<0.05);心俞穴穴区组织ATP显著降低(P<0.05),ADP降低,但差异无统计学意义(P>0.05),AMP浓度显著增加(P<0.05)。与急性心肌缺血组比较,预电针心俞穴组血清SOD含量显著增加(P<0.05);心俞穴穴区组织ATP、ADP含量升高,但差异无统计学意义(P>0.05),AMP含量显著降低(P<0.05)。结论预电针心俞穴对大鼠急性心肌缺血损伤具有一定的保护作用,心俞穴穴区组织中嘌呤物质可能启动了针刺信号的转换和起始传递。Objective To investigate the possible effect of acupoint local purine substance on improving acute myocardial ischemia in rats by pre-electroacupuncture at Xinshu point.Methods 18 male clean grade SD rats were randomly divided into saline control group,acute myocardial ischemia group and pre-electroacupuncture Xinshu point group,with 6 rats in each group.In the acute myocardial ischemia group,isoproterenol hydrochloride(ISO)was injected subcutaneously into the back at multiple points to prepare the rat model of acute myocardial ischemia.The pre-electroacupuncture Xinshu point group was treated with electroacupuncture at both Xinshu points for 3 d before modeling.The activity of creatine kinase MB isoenzyme(CK-MB)and superoxide dismutase(SOD)in serum were determined by automatic biochemical analyzer,and the concentrations of purine substance adenosine triphosphate(ATP),adenosine diphosphate(ADP)and adenosine monophosphate(AMP)in acupoint tissue of Xinshu acupoints were determined by enzyme-linked immunosorbent assay(ELISA).Results Compared with the saline control group,the content of serum SOD was significantly decreased in acute myocardial ischemia group(P<0.05),the content of ATP in acupoint tissue of Xinshu acupoints was significantly decreased(P<0.05),and ADP was decreased,but the difference was not statistically significant(P>0.05),while the content of AMP was significantly increased(P<0.05).Compared with the acute myocardial ischemia group,the content of serum SOD was significantly increased in the pre-electroacupuncture Xinshu point group(P<0.05),the contents of ATP and ADP in acupoint tissue of Xinshu point were increased,but the difference was not statistically significant(P>0.05),while the content of AMP was significantly decreased(P<0.05).Conclusions Pre-electroacupuncture at Xinshu point has a certain protective effect on acute myocardial ischemia injury in rats.Purines in acupoint tissue of Xinshu point might initiate the transformation and initial transmission of acupuncture signal.
