摘 要:目的:建立气阴两虚证氧化应激复合模型,基于证效相关探讨芝斛方的抗氧化作用及益气养阴功效。方法:采用单日限制饮食、双日负重力竭游泳,从第16天开始灌胃左甲状腺素钠片(150μg/kg)连续14 d的方式造气阴两虚证,实验最后一天灌胃50%的乙醇12 mL/kg造气阴两虚证复合氧化应激模型;观察小鼠的一般状态、检测小鼠体质量、力竭游泳时间、脏器指数,通过酶联免疫吸附法检测血清环磷酸腺苷(cAMP)、环磷酸鸟苷(cGMP)、三碘甲状腺原氨酸(T3)、四碘甲状腺原氨酸(T4)、免疫球蛋白M(IgM)、免疫球蛋白G(IgG)含量,采用比色法检测肌肉组织乳酸(LA)、肌糖原(MG)含量及肝脏组织Na^(+)-K^(+)-ATP酶、Ca^(2+)-Mg^(2+)-ATP酶、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)活性及丙二醛(MDA)含量,采用连续监测法测乳酸脱氢酶(LDH)、肌酸激酶(CK)活性变化。结果:与模型组比较,给药芝斛方的各组小鼠精神状态明显好转,芝斛方低、中、高剂量组第20、30天体质量增加(P<0.05,P<0.01),力竭游泳时间延长(P<0.01),胸腺指数显著增加(P<0.01),血清cAMP、cAMP/cGMP值显著降低(P<0.05,P<0.01),肌肉组织中LA含量显著减少(P<0.01),MG含量显著增加(P<0.01),肝脏组织Ca^(2+)-Mg^(2+)-ATP酶、SOD和GSH-Px活性显著增强(P<0.05,P<0.01),MDA含量显著减少(P<0.05);芝斛方低、高剂量组小鼠脾脏指数显著增加(P<0.05),芝斛方低、中剂量组CK活性显著增强(P<0.01),芝斛方高剂量组血清IgM含量显著升高(P<0.05),芝斛方中、高剂量组肝脏组织Na^(+)-K^(+)-ATP酶活性显著增强(P<0.01);芝斛方对小鼠血清T3、T4及IgG含量无显著性影响。结论:本研究基于辨证保健理论,建立了用于评价气阴两虚证氧化应激小鼠模型的方法,并阐释了芝斛方的益气养阴功效和抗氧化功能。Objective:To establish a composite model of oxidative stress of qi and yin deficiency syndrome,and to explore the replenishing qi and nourishing yin effect and antioxidant function of Zhihu Prescription.Methods:The model was induced by daily diet restriction,daily swimming with heavy load,and gavage of levothyroxine sodium tablets(150μg/kg)continuously for 14 days from the 16th day,and the mice were gavaged 50%ethanol(12 mL/kg)on the last day of the experiment.The general state,body weight,exhaustive swimming time and organ index of the mice were observed.The contents of cAMP,cGMP,T3,T4,IgM and IgG in serum of the mice were detected by Elisa.The contents of LA and MG,the activities of Na^(+)-K^(+)-ATP ase,Ca^(2+)-Mg^(2+)-ATP ase,SOD and GSH-Px and the content of MDA in liver were detected by colorimetry.the activities of LDH and CK in muscle were detected by continuous monitoring method.Results:Compared with model group,the mental state of mice treated with Zhihu Prescription was significantly improved.In all Zhihu Prescription groups,the body weight of mice at the 20th and 30th day was increased(P<0.05,P<0.01),the exhaustion swimming time was prolonged(P<0.01),the thymus index was increased(P<0.01),the serum cAMP and cAMP/cGMP levels decreased(P<0.05,P<0.01),the LA content in muscle tissue decreased(P<0.01),the MG content were significantly increased(P<0.01),the activities of Ca^(2+)-Mg^(2+)-ATP,SOD and GSH-Px in liver tissue were significantly increased(P<0.05,P<0.01),and the MDA content decreased significantly(P<0.05);the spleen index was increased in low dose and high dose Zhihu Prescription groups(P<0.05),the activity of CK was significantly increased in low dose and medium dose Zhihu Prescription groups(P<0.01),and the serum IgM content was increased in high dose Zhihu Prescription group(P<0.05),the activity of Na^(+)-K^(+)-ATP ase in liver tissues was increased in medium dose and high dose Zhihu Prescription groups(P<0.01);Zhihu Prescription had no significant effect on serum T3,T4 and IgG contents.Conclu
