摘 要:目的:探究热敏灸对腹泻型肠易激综合征(IBS-D)模型大鼠结肠脑源性神经营养因子(BDNF)和酪氨酸激酶受体B(TrkB)表达的影响,以及热敏灸治疗IBS-D的可能机制。方法:将42只雄性SD大鼠随机分为空白组,造模组。模型组成功建立IBS-D大鼠模型后随机分为模型组、7,8-DHF组、ANA-12组、艾灸组,各组根据分组方式分别予以对应干预措施,干预周期14 d后,将艾灸组大鼠根据尾温变化分为热敏灸组(≥1℃)及非热敏灸组(<1℃)。期间观察各组大鼠一般情况,HE染色观察结肠组织病理形态学变化,免疫组织化学法及免疫印迹法检测结肠BDNF和TrkB蛋白表达情况。结果:与模型组比较,热敏灸组可显著降低BDNF及TrkB蛋白表达(P<0.05,P<0.01)。结论:热敏灸可有效改善IBS-D大鼠胃肠动力异常,可能是通过调节BDNF、TrkB蛋白表达产生作用。Objective:To investigate the effects of heat-sensitive moxibustion on the expression of brain-derived neurotrophic factor(BDNF)and tyrosine kinase receptor B(TrkB)in the colon of rats with diarrhea-predominat irritable bowel syndrome(IBS-D),and the possible mechanism of heat-sensitive moxibustion in the treatment of IBS-D.Methods:Forty-two male SD rats were randomly divided into blank group and model group.Model group after successful,IBS-D rat model were randomly divided into model group,7,8-DHF group,ANA-12 group and moxibustion group,each corresponding intervention measure according to the grouping respectively intervention period for 14 days,and finally the moxibustion group rats when statistics according to end temperature change can be divided into heat-sensitive moxibustion group(≥1℃)and non-heat sensitive moxibustion group(<1℃).Finally,the general condition of rats in each group were observed,the pathological changes of colon tissue were observed by HE staining,and the protein expressions of BDNF and TrkB in colon were detected by immunohistochemistry and Western Blot.Results:Compared with the model group,the heat-sensitive moxibustion group could significantly reduce the protein expressions of BDNF and TRKB(P<0.05,P<0.01).Conclusion:Heat-sensitive moxibustion can effectively improve gastrointestinal motility in IBS-D rats,possibly by regulating the expression of BDNF and TrkB proteins.
