摘  要:研究交趾黄檀心材对冠脉左前降支结扎大鼠血浆内源性代谢物的影响,分析交趾黄檀心材改善急性心肌缺血损伤的作用机制。通过指纹图谱的建立验证交趾黄檀心材成分的稳定一致后,将雄性SD大鼠30只,随机分为假手术组、模型组、交趾黄檀心材组(6 g·kg^(-1)),每组10只。假手术组只开胸穿线不结扎,其余组构建冠脉左前降支结扎大鼠模型。给药10 d后取材取血,进行心脏HE染色;检测血浆中心肌损伤指标肌酸激酶同工酶(creatine kinase isoenzyme,CK-MB)、乳酸脱氢酶(lactate dehydrogenase,LDH)的含量,能量代谢相关指标葡萄糖(glucose,Glu)以及血管内皮功能指标一氧化氮(nitric oxide,NO)的含量;并采用高效液相-飞行时间-质谱联用技术检测血浆内源性代谢物。结果表明,交趾黄檀心材能降低模型大鼠血浆中CK-MB及LDH含量,减轻心肌损伤;血浆中Glu含量降低,提示心肌能量代谢的改善;NO含量升高,血管内皮损伤改善,促进血管扩张。交趾黄檀心材可以改善大鼠冠脉左前降支结扎导致的细胞间隙增大、心肌炎性细胞浸润及肌丝断裂现象。代谢组学研究显示,模型组大鼠血浆中26种代谢产物含量显著上升,27种代谢产物含量显著下降;给予交趾黄檀心材后能明显回调其中20种代谢产物含量。交趾黄檀心材能够显著回调因冠脉左前降支结扎的大鼠代谢异常,其机制可能是调节心脏能量代谢、NO生成以及炎症等途径。该结果可为进一步解释交趾黄檀改善急性心肌损伤作用提供相应依据。This paper aimed to study the effect of Dalbergia cochinchinensis heartwood on plasma endogenous metabolites in rats with ligation of the left anterior descending coronary artery, and to analyze the mechanism of D. cochinchinensis heartwood in improving acute myocardial ischemic injury. The stability and consistency of the components in the D. cochinchinensis heartwood were verified by the establishment of fingerprint, and 30 male SD rats were randomly divided into a sham group, a model group, and a D. cochinchinensis heartwood(6 g·kg^(-1)) group, with 10 rats in each group. The sham group only opened the chest without ligation, while the other groups established the model of ligation. Ten days after administration, the hearts were taken for hematoxylin-eosin(HE) staining, and the content of heart injury indexes in the plasma creatine kinase isoenzyme(CK-MB) and lactate dehydrogenase(LDH), energy metabolism-related index glucose(Glu) content, and vascular endothelial function index nitric oxide(NO) was determined. The endogenous metabolites were detected by ultra-high-performance liquid chromatography-time-of-flight-mass spectrometry(UPLC-Q-TOF-MS). The results showed that the D. cochinchinensis heartwood reduced the content of CK-MB and LDH in the plasma of rats to relieve myocardial injury, reduced the content of Glu in the plasma, improved myocardial energy metabolism, increased the content of NO, cured the vascular endothelial injury, and promoted vasodilation. D. cochinchinensis heartwood improved the increase of intercellular space, myocardial inflammatory cell infiltration, and myofilament rupture caused by ligation of the left anterior descending coronary artery. The metabolomic study showed that the content of 26 metabolites in the plasma of rats in the model group increased significantly, while the content of 27 metabolites decreased significantly.
