摘  要:目的分析湖北省麻疹病毒流行株基因型特征。方法收集湖北省2016-2021年麻疹监测病例咽拭子标本,进行麻疹病毒核酸检测、分离培养、N基因C末端基因扩增,构建分离株与基因库(GenBank)中麻疹病毒基因型参考株的系统进化树,分析核苷酸同源性和基因型别。结果从湖北省2016-2021年麻疹监测病例中共获得335株麻疹病毒流行株,其中H1基因型327株(2016-2018年)、B3基因型5株(2019年)、疫苗株A基因型3株(2016年和2021年)。H1基因型麻疹病毒分离株之间的核苷酸同源性为98.40%-98.90%,与H1基因型参考株的核苷酸同源性为97.30%-97.80%;B3基因型麻疹病毒分离株之间的核苷酸同源性为99.30%-100%,与B3基因型参考株的核苷酸同源性为97.10%-97.60%。结论2016-2018年湖北省麻疹病毒优势流行株仍为中国本土H1基因型,2019年首次发现B3基因型,2021年仅检测到疫苗株A基因型。需继续加强麻疹病毒基因型监测,及时发现和阻断输入性麻疹传播。Objective To analyze genetic characteristics of measles virus(MV)isolates in Hubei province.Methods We collected throat swabs from measles cases reported between 2016 and 2021 in Hubei province for MV nucleic acid detection,isolation and culture,and amplification of the C-terminal N gene.We constructed a phylogenetic tree of the MV isolates with GenBank MV genotype reference strains to determine nucleotide homologies and genotypes.Results A total of 335 MV strains were obtained from measles surveillance cases from 2016 to 2021 in Hubei,among which 327 were H1 genotype(in 2016-2018),5 were B3 genotype(in 2019),and 3 were vaccine strain A genotype(in 2016 and 2021).Nucleotide homologies within H1 genotype isolates were 98.40%-98.90%and were 97.30%-97.80%between H1 genotype isolates and reference strains;homologies within B3 genotype isolates were 99.30%-100%and were 97.10%-97.60%between B3 genotype isolates and reference strains.Conclusions The predominant MV strains from 2016 to 2018 in Hubei were indigenous H1 genotype MVs.B3 genotype MVs were found in 2019,but only A genotype was found in 2021.We should strengthen monitoring MV genotypes for rapid identification and interruption of imported measles transmission.
