摘 要:目的将雪旺细胞(SCs)和血管内皮细胞以不同比例体外共培养,在实验中找到培养最佳比例,为体内构建血管化组织工程神经提供实验基础。方法从大鼠坐骨神经及胸主动脉分离、培养雪旺细胞和血管内皮细胞,设雪旺细胞单独培养为对照组A,以1∶1、1∶4、4∶1、8∶1、1∶8的比例联合共培养为B、C、D、E、F实验组。在共培养的第1、3、5和7天,对雪旺细胞计数,绘制其生长曲线,分析细胞共培养的实验结果。结果在倒置显微镜下观察,每组SCs数量随着培养时间出现不同的变化,在共培养第1天,各实验组与对照组A细胞数量差异无统计学意义(P>0.05),从共培养的第3天至第7天,实验组B(1∶1)、C(1∶4)和F(1∶8)细胞计数与对照组A相比差异有统计学意义(P<0.05),实验组D(4∶1)、E(8∶1)与对照组A的差异无统计学意义(P>0.05),但与其他实验组B、C、F相比差异具有显著的统计学意义(P<0.05),实验组D与实验组E比较差异有统计学意义(P<0.05)。结论通过本实验的步骤可获取原代雪旺细胞和血管内皮细胞,实验统计分析得出雪旺细胞和血管内皮细胞共培养最适比例为4:1,为组织工程神经的血管化实验提供了理论基础。Objective Co-cultivate schwann cells and vascular endothelial cells in different proportions in vitro,find the best proportion in the experiment,and provide experimental basis for the construction of vascularized tissue engineering nerve in vivo.Methods Schwann cells and vascular endothelial cells were isolated and cultured from rat sciatic nerve and thoracic aorta.Schwann cells were separately cultured as control group A,and co-cultured with the ratio of 1:1,1:4,4:1,8:1,1:8 into experimental groups B,C,D,E,and F.On the 1st,3rd,5th and 7th days of co-culture,Schwann cells were counted,their growth curves were drawn,and the experimental results of co-culture were analyzed.Results Under the inverted microscope,the number of SCs in each group varied with the culture time.On the first day of co-culture,there was no significant difference in the number of A cells between the experimental groups and the control group(P>0.05).From the third day to the seventh day of co-culture,the number of B(1:l),C(1:4)and F(1:8)cells in the experimental group was significantly different from that in the control group A(P<0.05),and the number of D(4:l)and E(8:1)in experimental group has no significant difference with control group A(P>0.05),but they have significant difference with other experimental groups B,C and F(P<0.05).There was significant difference between experimental group D and E(P<0.05).Conclusion Primary Schwann cells and vascular endothelial cells can be obtained through the steps of this experiment.The experimental statistical analysis shows that the optimal ratio of Schwann cells and vascular endothelial cells is 4:1,which provides a theoretical basis for the vascularization experiment of tissue engineering nerve.
