摘 要:天然海洋生物黏附蛋白分泌量极少、直接提取受到技术限制,难以满足日益增长的市场需求。借助于基因工程手段,近些年发展起来的海洋生物黏附蛋白体外表达、纯化和检测技术为上述难题提供了解决思路,由此制备的具有生物活性的黏附蛋白在实际应用过程中表现出色。本文全面综述了海洋生物黏附蛋白体外制备的研究进展,主要集中在蛋白表达系统、蛋白纯化体系和蛋白检测技术等方面。同时,基于现有进展对未来相关技术的发展趋势也进行了展望,以期推动海洋生物黏附蛋白研究和应用工作广泛开展,为解决海洋生态环境问题、促进海洋生物来源的仿生材料研发提供技术支撑。Marine organisms usually secret little natural adhesive proteins.The direct extraction of these proteins is limited by our recent technologies,which makes it difficult to meet the growing market demand.With the help of genetic engineering,the technologies in vitro expression,purification and detection of marine adhesive proteins have been rapidly developed in recent years,providing a solution to the above problems.This paper comprehensively reviews the research progress of in vitro preparation of marine adhesive proteins,mainly focusing on the protein expression system,protein purification system and protein detection technology.Meanwhile,based on the existing research progress,the developmental trend of related technologies in the future is prospected to promote the extensive investigation and application of marine adhesive proteins,and provide enough technical support for solving marine ecological environment issues and promoting the development of biomimetic materials derived from marine adhesive organisms.
